海棠叶根皮苷药理活性的研究 被引量:4

Pharmacological activity of phlorizin extracted from leaves of Malus hupehensis

摘要 为了研究海棠叶提取物根皮苷的药理活性,以本实验室提取的根皮苷为试材,利用清除自由基检测体系测定根皮苷的抗氧化活性;并以卵巢癌细胞(Skov3)为细胞模型,检测根皮苷的抗肿瘤活性。结果显示:根皮苷对各种自由基具有一定的清除作用,清除效果依次为2,2'-连氮基-双-(3-乙基苯并二氢噻唑啉-6-磺酸)二铵盐(ABTS)自由基〉二苯代苦味酰基(DPPH)自由基〉羟基自由基〉金属离子Fe2+自由基,且具有一定的浓度依赖性;根皮苷可引起卵巢癌细胞(Skov3)细胞内活化氧(ROS)的表达量的增加,造成核DNA的泄露,并诱导细胞的凋亡。结果表明:根皮苷具有抗氧化作用和抗肿瘤活性,此为根皮苷用于肿瘤的治疗奠定了良好的基础。 To explore the antioxidant and antitumor activity of phloridzin extracted from leaves of Malus hupehensis,the samples of phloridzin extracted and preserved in our lab were used to determine its anti-oxidative activity via the free radical detection system. And the Skov3 cell was used as a model to measure the antitumor activity of phlorizin. The antioxidant activity of phloridzin was realized by Diphenyl bitter acyl radical,DPPH radical,hydroxy radical,Fe2+radical and Az BTS-(NH4) 2(ABTS) radical. Results showed that phloridzin was able to clean four radicals and the scavenging effect on the radicals was in the order of ABTS radical DPPH radical the hydroxyl radicalFe2+radical. Compared with the control group,the fluorescence intensity of ROS was significantly higher in the treated group of Skov3 cells;and phloridzin caused nuclear DNA leakage,leading to the eventual celluar apoptosis,which implies that phloridzin had antitumor activity.This study provides basic data for the widespread use of phlorizin in antitumor research and treatment.
