摘要
目的检测microRNA-10b(miR-10b)在大肠癌患者中的表达情况,并探讨miR-10b对大肠癌SW480细胞侵袭和转移的影响。方法收集大肠癌患者标本32例,同时收取癌组织及相应的癌旁组织。通过荧光定量PCR(qRT-PCR)方法检测各组织中miR-10b的表达水平。以脂质体为载体将miR-10b抑制剂转染至SW480细胞,Transwell实验检测转染后SW480细胞的侵袭和迁移能力变化,利用qRT-PCR和免疫印迹实验检测Kriippel样因子4(KLF4)mRNA和蛋白水平的改变。结果miR-10b在32例患者癌组织及正常黏膜组织中的表达量分别为(0.24±0.09)与(0.67±0.28),差异有统计学意义(P<0.01)。下调miR-10b的表达能够显著抑制SW480细胞的侵袭和迁移能力,KLF4 mRNA和蛋白的表达水平均显著升高,P<0.01。结论MicroRNA-10b在大肠癌中低表达,并可能通过上调KLF4的表达抑制大肠癌SW480细胞侵袭和转移。
Objective To examine the expression of micro RNA-10b(mi R-10b)in colorectal cancer,and to investigate the effects of mi R-10b on invasion and migration of SW480 cell line.Methods We detected mi R-10b expressions by quantitative real-time PCR(q RT-PCR)in 32 colorectal cancer tissues and their corresponding adjacent tissues.The mi R-10b inhibitor was transfected into SW480 cells with Lipofectamine,and invasion and migration ability of SW480 cells were detected by transwell cell assay,q RT-PCR and Western blot were used to examine the m RNA and protein expression of Kriippel like factor 4(KLF4)respectively.Results The expression level of mi R-10b in the carcinoma tissues and normal tissues of 32 patients were(0.24±0.09)and(0.67±0.28)respectively,and there was significant difference statistically(P〈0.01).The invasion and migration ability of SW480 cells were markedly inhibited by the inhibition of mi R-10b,and the expression of KLF4 m RNA and protein was upregulated significantly too(P〈0.01).Conclusion The expression of mi R-10b in colorectal cancer tissues was down-regulated,and the inhibition effect of mi R-10b inhibitor on the invasion and migration ability of SW480 cells might be related to the upregulation of KLF4 expression.
