摘要
目的探讨番茄红素对PS1/APP小鼠前额叶皮质区Notch1、GRP78表达及学习记忆功能的影响。方法将18只6月龄的PS1/APP小鼠随机平分为3组,分别为正常对照组、模型组、模型组+番茄红素组;利用Morris水迷宫测试小鼠学习记忆功能,免疫组化、Western blot检测Notch1和GRP78在小鼠前额叶皮质区的表达情况。结果番茄红素明显改善PS1/APP小鼠的空间学习记忆功能,下调前额叶皮质区Notch1和GRP78的表达水平。结论番茄红素改善PS1/APP小鼠学习记忆功能可能与下调前额叶皮质区Notch1和GRP78蛋白的表达水平相关。
Objective To investigate the effect of lycopene on Notch1 and GRP78 expression in the prefrontal cortex and learning and memory abilities of PS1/APP mice.Methods Eighteen 6-month-old PS1/APP mice were randomly divided into three groups:normal control group,model group and model+lycopene group.Morris water maze test was used to study the memory function.Immunohistochemistry and Western blot were used to detect Notch1 and GRP78expression in the prefrontal cortex.Results Lycopene significantly improved the spatial learning and memory abilities and downregulated the expression of Notch1 and GRP78 in the prefrontal cortex of PS1/APP mice.Conclusion The improvement of learning and memory abilities by lycopene in PS1/APP mice might be related to the downregulation of Notch1 and GRP78 proteins expression in prefrontal cortex.
