摘要
目的研究微小核糖核酸29(miRNA29)靶基因细胞周期素D2重组蛋白(CCND2)及周期蛋白依赖性激酶6(CDK6)在宫颈鳞癌组织中的表达及其与临床病理各因素之间的关系。方法采用逆转录聚合酶链式反应(RT-PCR)检测宫颈鳞状细胞癌组织、子宫颈上皮内瘤变(CIN)组织、正常宫颈组织中CCND2、CDK6的信使核糖核酸(mRNA)水平。结果miRNA29靶基因CCND2在正常宫颈组织、CIN及宫颈鳞状细胞癌组织中的表达有统计学差异(H=29.27,P=0.00)。miRNA29靶基因CDK6在宫颈鳞状细胞癌组织、CIN及正常宫颈组织中表达水平无统计学差异(H=2.76,P=0.25)。在CIN组织中,CCND2与CDK6存在正相关(r=0.58,P<0.05)。结论 miRNA29靶基因CCND2可能参与宫颈癌的发生发展;CDK6与宫颈癌发生发展缺乏相关性;CCND2和CDK6在CIN发生发展中可能起协同作用。
Objective To study the expressions of recombinant cyclin D2(CCND2)and cyclin-dependent kinases 6(CDK6)in miR-29 target gene and their relationships with clinicopathology.Methods The mRNA in CCND2 and CDK6 was detected in normal cervical tissues,CIN tissues and cervical squamous cell carcinoma tissues by reverse transcription-polymerase chain reaction(RTPCR).Results There was significant difference in CCND2 expression among normal cervical tissues,CIN tissues and cervical squamous cell carcinoma tissues(H =29.27,P=0.00),but there was no significant difference in CDK6 expression among them(H =2.76,P=0.25).CCND2 was positively correlated with CDK6 in CIN tissue(r=0.58,P0.05).Conclusion MiR-29 target gene CCND2 may be involved in the occurrence and development of cervical cancer,but CDK6 is not related to the occurrence and development of cervical cancer.CCND2 and CDK6 may play a synergistic role in CIN genesis and development.
