摘要 为更好的开发与利用甘薯资源,以甘薯为原料,采用水提醇沉的方法从甘薯中提取多糖并进行含量测定.通过单因素试验与正交试验对甘薯多糖成分提取工艺进行优化设计,以期确定甘薯多糖的最佳提取工艺.由试验结果可知,葡萄糖标准曲线的回归方程为Y=12.886X+0.001 5,r=0.9998,n=6.在0.009 94~0.059 64 mg/mL质量浓度范围内呈良好的线性关系.结果显示,其最优的提取工艺条件是:提取时间为5 h、提取温度为90℃、料液比为1∶15(m/V).在该工艺条件下,甘薯多糖的提取率为1.78%. For better development and utilization of sweet potato resources,in this paper,with sweet potato as raw material,polysaccharides were extracted from sweet potato using water extract-alcohol precipitation,and the content was determined. The single factor test and orthogonal test were applied to optimize sweet potato polysaccharide extraction process aiming to determine the sweet potato best extraction technology of polysaccharides. The test result showed that glucose standard curve regression equation was Y = 12. 886 X + 0. 001 5,r =0. 999 8,n = 6. The concentration had a good linear relationship within the scope of 0. 00994 ~ 0. 059 64 mg/mL. Results showed that the optimal extraction process conditions were as follows: extraction time of 5 h,extraction temperature of 90 ℃,material liquid ratio of 1:15( m/V). Under the condition of the process,sweet potato extraction yield of polysaccharide was 1. 78%.
