摘要
香蕉与枯萎病互作机理研究一直是近年来研究的热点,本文从香蕉根部克隆到一个lncRNA基因MalncPSA,通过生物信息学发现其位于异分枝酸合成酶基因ICS上游2 605 bp处。通过RT-PCR检测,对这2个基因在感、抗病香蕉种质中的表达水平进行分析,结果表明,MalncPSA和MaICS的表达趋势相似,在枯萎病菌侵染香蕉植株后3-27 h表达水平增加,而到51 h表达水平下降,并且在抗性种质中的变化幅度比在感病种质中大。对香蕉根部水杨酸的测定也表明,水杨酸含量的变化与这2个基因表达水平的变化相似。本研究结果表明,MalncPSA可能参与香蕉受胁迫下水杨酸的合成调控。
The interaction between banana plant and Fusarium oxysporum f. sp. cubense has brought more and more attentions in recent years. The lncRNA gene MalncPSA was cloned from the banana roots, which exists in the 2605 bp upstream of isochorismate synthase (ICS) gene. The expression levels of MalncPSA and MalCS showed very similar trend in banana roots after inoculation by F. oxysporum. Their expression levels increased from 3 h to 27 h after inoculation and then decreased at 51 h. In addition, the fold changes of their expression levels were higher in the disease-resistant banana than in the disease-sensitive banana. The concentration of salicylic acid (SA) was similar with the fold change of the two genes in two banana cultivars. The research indicated that MalncP- SA might be involved in the biosynthesis of SA in banana under the stress.
