丁苯酞注射液对大鼠脑出血后血肿周围组织AQP4表达的影响 被引量:1

摘要 目的观察丁苯酞注射液对大鼠脑出血后血肿周围组织水通道蛋白4(AQP4)表达的影响。方法将SD雄性大鼠55只随机分为假手术对照组(n=5)、脑出血组(n=25)、丁苯酞治疗组(n=25),采用自体血注入法建立脑出血大鼠模型,成功后各时间点分别行干—湿重法计算脑组织含水量、免疫组化测定AQP4的表达情况。结果与假手术对照组相比较,脑出血组在6 h后AQP4及脑组织含水量开始增加,有显著差异(P<0.05),3 d达到高峰(P<0.05),之后开始下降,到7 d仍未恢复正常。丁苯酞治疗组各时间点AQP4及脑组织含水量均显著低于脑出血组(P<0.05)。结论丁苯酞注射液可通过影响AQP4的表达而改善脑出血后脑水肿而达到保护作用。 Objective To investigate the effect of butylphthalide on expression of AQP4 around the hematoma in experimental intracerebral hemorrhage( ICH). Methods 55 SD rats were randomly divided into sham operated control group( n = 5),ICH group( n = 25) and butylphthalide treatment group( n = 25). The intracerebral hemorrhage model was formed by infusing autologous artery blood into caudate nucleus. The changes of brain water content were measured by the wet and dry method at different time point after ICH. Immunohistochemistry straining was used to detect the changes of expression of AQP4.Results Compared with the sham operation control group,the expression of AQP4 increased in ICH group at 6 h( P 0. 05),reached a peak at 3 d( P 0. 05),then began to decline,and was still higher than normal at 7 d. The expression of AQP4 were significantly lower in butylphthalide treatment group than in ICH group each time point( P 0. 05). Conclusion Butylphthalide injection can alleviate cerebral edema by regulate AQP4 expression in rats following intracerebral hemorrhage.
