摘要
为了提高海带甘露醇的浸提率,以海带为原料,用果胶酶酶解浸提甘露醇。在单因素试验的基础上,选用响应面法进行优化分析,最后确定果胶酶酶解浸提海带甘露醇的最佳工艺条件为:果胶酶添加量0.26 m L/L、料液比1∶39 g/m L、浸提时间59 min、浸提温度52℃。在此条件下,海带甘露醇达到最高获得率,为6.19%,与预测值的相对误差为1.98%。试验验证了响应面法对于寻找最佳浸提条件的可行性。
This study was designed to improve the extraction rate of mannitol from Laminariajaponica Areseh by applying pectinase enzymolysis extraction. In our study, the Laminaria japonica Areseh was applied as the raw material, then series of control experiments were set to analyze the sensitivities of the following experimental factors which were the dose of pectinase, solid-liquid ratio, soaking time and extraction temperature. Each experiment factor was analyzed by single-factor method, and response surface methodology was also used for optimize analysis. The experiment results showed a group of optimum extraction conditions for the maximum extraction rate of mannitol from Laminariajaponica Areseh were the dose of pectinase of 0.26 mL/L, solid-liquid ratio of 1 : 39 g/mL, soaking time of 59 min and extraction temperature of 52 ℃. Under the optimized conditions, the yield of mannitol from Laminariajaponica Areseh was 6.19%. The relative error was 1.98%. The study confirmed the feasibility of applying the response surface methodology in looking for the optimum extraction conditions of the maximum extraction rate ofmannitol from Laminariajaponica Areseh.
