姜黄素联合热疗对人黑素瘤A375细胞毒性的实验研究

摘要 目的从细胞水平探讨姜黄素联合热疗治疗人恶性黑素瘤的可行性。方法采用噻唑蓝(MTT)比色法检测不同处理因素对人恶性黑素瘤A375细胞增殖活性的影响,流式细胞仪检测A375细胞的凋亡情况。结果 30μmol/L姜黄素联合热疗的细胞抑制率高于单纯姜黄素组和热疗组,差异具有统计学意义(F=5.47,P<0.01);A375细胞增殖抑制率与姜黄素浓度呈正相关(rs=0.95);30μmol/L姜黄素联合热疗组细胞早期凋亡率高于单纯姜黄素组和热疗组,差异具有统计学意义(H=15.37,P<0.01)。结论姜黄素联合热疗对人恶性黑素瘤A375细胞的毒性起协同相加作用,细胞毒性与姜黄素的剂量成明显剂量-效应关系。 Objective To investigate the feasibility of applying curcumin combined with thermotherapy in the treatment of malignant melanoma at the cell level. Methods MTT was used to detect the effects of different treatment factors on cell growth of human malignant melanoma A375 cells, and the apoptosis of A375 cells was detected by flow cytometry. Results The inhibition of 30 μmol/L curcumin combined with thermotherapy was higher than that of simple curcumin group and thermotherapy group, the difference was statistically significant(F=5.47, P〈0.01). The inhibition rate of A375 cells was correlated with the concentration of curcumin(rs=0.95); the apoptosis rate of 30 μmol/L curcumin combined with thermotherapy group was higher than the simple curcumin group and thermotherapy group, the difference was statistically significant(H=15.37, P〈0.01). Conclusion Curcumin combined with thermotherapy has synergistic effects on the cytotoxicity of human malignant melanoma A375 cells, and the cytotoxicity and the dose of curcumin have obvious dose effect relationship.
