Celastrol specifically inhibits the activation of NLRP3 inflammasome

摘要 Dear Editor, Inflammasomes are vital components of the innate immune system that survey microbial infections or sterile challenges in the cytosol (de Zoete et al., 2014). A typical inflamma- some is composed of a sensor protein such as NOD like receptor (NLR) or AIM2 like receptor (ALR), the adaptor protein apoptosis associated speck-forming complex con- taining CARD (ASC), and the effector protein pro-caspase-1 (Vanaja et al., 2015). Assembly of inftammasome leads to the maturation of caspase-1, which cleaves pro-IL-l[3 and pro-IL-18 to their mature forms, and results in pyroptosis (Shi et al., 2017). A number of infiammasomes have been identified so far, and the NLRP3 inflammasome is the most extensively studied one due to its connection to many dif- ferent diseases (Guo et al., 2015). As such, more and more chemical inhibitors have been developed to control the func- tion of NLRP3 inflammasome (Shao et al., 2015).
