Estrogenic Effect of Erzhiwan on Immature Mice

摘要 目的:观察二至丸对未成熟小鼠子宫系数、血清中雌二醇(estrodiol,E2),子宫阴道形态学变化及雌激素受体(estrogen receptor,ER)α和ERβ表达的影响,探讨其发挥雌激素样作用机制。方法:取健康,出生21 d的昆明种雌性小鼠40只,随机分成4组,分别为空白组,阳性药戊酸雌二醇(EV)组(0.154 mg·kg^-1),二至丸1.365,2.73 g·kg^-1组,每组10只,连续给药7 d后观察小鼠子宫系数;分别采用酶联免疫吸附法(ELISA)检测血清中E2的水平;苏木素-伊红(HE)染色法观察未成熟小鼠子宫阴道形态改变;蛋白免疫印迹法(Western blot)检测子宫、阴道ERα和ERβ的蛋白表达水平。结果:与空白组比较,二至丸1.365 g·kg^-1组能使未成熟小鼠子宫系数显著增加(P〈0.05),血清中E2含量明显升高(P〈0.05),能明显促进靶器官子宫、阴道的生长发育,还能显著上调阴道ERα和ERβ表达(P〈0.05),与EV作用相似。结论:二至丸通过上调血中雌二醇含量和靶器官雌激素受体表达发挥雌激素样作用。 Objective: To observe the effect of Erzhiwan on uterus coefficient,estrodiol(E2) in serum,pathological changes in tissues of uterus and vagina,and expressions of estrogen receptors(ERα and ERβ) of immature mice,in order to investigate the estrogen-like mechanism,and provide experimental evidence for the clinical application of Erzhiwan in the treatment of perimenopausal syndrome.Method: Forty female Kunming mice were randomly divided into 4 groups: normal group,estrogen receptor agonist group(EV,0.154 mg·kg^-1) and Erzhiwan(1.365,2.73 g·kg^-1) groups.Seven days later,the serum level of E2 was detected by enzyme-linked immunosorbent assay(ELISA).The pathological changes in immature mouse tissues were observed by hematoxylineosin(HE) staining.The expressions of ERα and ERβ were detected by Western blot.Result: Compared with normal group,Erzhiwan groups showed a significant increase in the uterus coefficient(P〈0.05) and a decrease in the level of E2(P〈0.05),and the growth and development of uterus and vagina,and increase of the expressions of ERα and ERβ in vagina of immature mice,with a similar effect with EV.Conclusion: Erzhiwan plays an estrogen-like effect by up-regulating the expression of estradiol in blood and the expressions of estrogen receptors in target organ.
