摘要 目的观察葛根通络颗粒的镇痛作用及其机制。方法将48只筛选合格小鼠随机分为:模型组、葛根通络颗粒低、中、高剂量组(3.0、6.0、12.0g·kg-1,以生药计,以下简称GG低组、GG中组、GG高组),模型组仅给予0.5%CMC-Na混悬液(0.2m L/10g),采用热板法观察各组小鼠痛阈值变化情况;将50只筛选合格小鼠随机分为模型组、GG低组、GG中组、GG高组和阿司匹林组(0.2g/kg),模型组仅给予0.5%CMC-Na混悬液(0.2m L/10g),各组小鼠均在给药后1小时按0.1m L/10g腹腔注射0.6%冰醋酸(HAC),观察15min后各组小鼠扭体反应次数,记录次数并计算抑制率;将50只筛选合格小鼠随机分为模型组、GG低组、GG中组、GG高组和空白对照组,除空白对照组外,其余四组各鼠均皮下注射利血平1.0mg/kg,连续注射10天,观察葛根通络颗粒对小鼠脑内5-HT含量的影响。结果痛阈值:给药前及给药0.5h后四组小鼠痛阈值无统计学意义(P≥0.05);GG高组给药后1h、1.5h、2h与模型组相比,痛阈值明显提高,差异有统计学意义(分别为P<0.01,P<0.01,P<0.05);GG中组给药后1.5h、2h与模型组相比,痛阈值明显提高,差异有统计学意义(P<0.05);GG低组给药后各个时间点与模型组相比痛阈值差异无统计学意义(P≥0.05)。扭体次数:GG低、中、高剂量组、阿司匹林组与模型组相比,都能显著减少冰醋酸导致小鼠扭体次数,差异有统计学意义(P<0.05)。脑内5-HT含量:模型组小鼠脑内递质5-HT较正常小鼠明显降低(P<0.05),GG高组对小鼠脑内的递质5-HT有明显的升高作用,与模型组比较差异有统计学意义(P<0.05),GG中、GG低组5-HT的含量较模型组也有一定程度的升高,但差异无统计学意义(P≥0.05)。结论葛根通络颗粒有一定的镇痛作用,机制可能与升高脑内5-羟色胺水平有关。 Objective: To observe the analgesic effect and the mechanism of pueraria Tongluo granules. Method: 48 eligible mice were randomly divided into: model group,puerarin Tongluo low,medium and high dose groups( 3. 0,6. 0,12. 0 g · kg-1),in terms of crude drug,hereinafter referred to as GG low group,GG Group,GG high group). Themodel group was treated with 0. 5% CMC-Na suspension( 0. 2 mL/10 g),and hot plate method was used to observe the change of pain threshold of mice. 50 eligible mice were randomly divided into model group,GG low group,GG Group,GG high group and aspirin group( 0. 2 g/kg). 0. 5% CMC-Na suspension( 0. 2 mL/10 g) was given to the model group.The mice in each group were injected intraperitoneally with 0. 6% acetic acid( HAC) at 0. 1 mL/10 g one hour after the administration. Mice writhing reaction times were recorded and the inhibition rate was calculated. 50 eligible mice were randomly divided into model group,GG low group,GG group,GG high group and blank control group,except the blank control group,the other four groups were subcutaneously injected reserpine 1. 0 mg/kg for continuous 10 days,the effect of pueraria granules on 5-HT content in mouse brain was observed. Result: Pain threshold: the pain thresholds of the four groups of mice before treatment and 0. 5 h after administration were not statistically significant( P≥0. 05). Compared with the model group,the pain threshold of the GG high group was significantly increased at 1 h,1. 5 h and 2 h( P〈0. 01,P〈0. 01,P〈0. 05,respectively). Compared with the model group,the pain threshold of the GG group was significantly increased at 1. 5 h and 2 h after administration( P〈0. 05),the difference was statistically significant( P〈0. 05).Compared with the model group at each time point,the difference of pain threshold of the GG low group was not statistically significant( P≥0. 05). Writhing times: Compared with the model group,the GG low,medium and high dose group and aspirin group could significantly reduce the writhing times of mice induced by glacial acetic acid,and the difference was statistically significant( P〈0. 05). 5-HT content in brain: The neurotransmitter 5-HT in the mice of the model group was significantly lower than that in the normal mice( P〈0. 05),GG high group showed a significant increase in 5-DT and the difference had statistical significance( P〈0. 05) compared with the model group,5-DT contents in GG and GG high groups were also increased to some extent while the difference had no statistical significance( P≥0. 05).Conclusion: pueraria Tongluo granules have a certain analgesic effect,and the mechanism may be related to the elevation of 5-HT level in the brain.
