摘要
目的:观察补肾活血汤对D-半乳糖致腰椎间盘退变大鼠模型IL-1β、TGF-β1表达的影响。方法:将72只健康SD大鼠随机分为空白组、模型对照组、模型组、中药组,每组18只,采用皮下注射D-半乳糖方法将模型组和中药组建立衰老大鼠椎间盘退变模型,模型对照组皮下注射等量生理盐水,在验证造模成功后,中药组灌服补肾活血汤药液,其余各组灌服等量生理盐水,灌胃4周后处死。观察比较各组腰椎间盘形态组织学和IL-1β、TGF-β1含量的变化。结果:模型组腰椎间盘组织退变程度评分、IL-1β、TGF-β1表达均高于空白组,差异均有统计学意义(P<0.05);中药组腰椎间盘组织退变程度评分、IL-1β、TGF-β1表达均低于模型组,差异均有统计学意义(P<0.05)。结论:补肾活血汤可降低D-半乳糖致腰椎间盘退变大鼠模型IL-1β、TGF-β1表达,延缓大鼠椎间盘的退变。
Objective： To observe the Effects of Bushen Huoxue Decoction on Expression of IL-1β and TGF-β1 of D-galactose lead to the intervertebral disc degeneration in rat model. Methods： 72 healthy SD rats were randomly divided into blank group, model control group, model group, Chinese medicine group, 18 rats in each group. The degeneration model of the intervertebral disc in aging rats was established by subcutaneous injection of D-galactose for the model group and the Chinese medicine group, while the model control group sub- cutaneous injection amount of normal saline. The Chinese traditional medicine group was dosed with Bushen Huoxue Decoction, and the rest of the groups with same volume of saline. All rats were killed after 4 weeks perfusion. Changes of histopathology of lumbar intervertebral disc and the contents of IL-1 beta and TGF- beta 1 were observed and compared between each group. Results： The model group showed higher degree of degeneration of lumbar intervertebral disc tissue and expression of IL-1, TGF-β1 than blank group. The difference was statistical significance （P〈0.05）. The Chinese medicine group showed lower degree of degeneration of lumbar in- tervertebral disc tissue and expression of IL-1, TGF-β1 than model group. The difference was statistical signifi- cance （P〈0.05）. Conclusion： Bushen Huoxue Decoction could lower the expression of IL-1β and TGF-β1 of Dgalactose lead to the intervertebral disc degeneration in rats model, and delay the degeneration of intervertebral discs of rats.
