摘要
目的:筛选半夏多糖除蛋白的最优工艺。方法:以半夏多糖保留率和蛋白脱除率为考察指标,研究3种脱蛋白方法对半夏多糖脱蛋白的影响,并用正交试验优选出最佳工艺。结果:确定了胰蛋白酶结合三氯乙酸的脱蛋白方法,最优脱蛋白工艺为:在胰蛋白酶最适酶解条件下酶解,酶解液与TCA溶液(5%)的体积比为4:1,振摇时间10 min,静置时间30 min;在此条件下半夏多糖蛋白脱除率为94.96%,多糖保留率为91.51%。结论:优选出的脱蛋白方法可以有效的去除蛋白,纯化多糖,为半夏多糖制备工艺的进一步研究提供了参考。
Objective: To screen the optimum method for deproteinization of Pinellia Ternate polysaccharides. Methods: Three deproteinization methods were evaluated in terms of the ratio protein removing and polysaccharide remaining by using orthogonal test to select the best technology. Results: Trypsin-trichloroacetic acid method was the best one,in which deproteination rate was94. 96% and 91. 51% of polysaccharide was remained. The optimum conditions for deproteinization were as follows. Under the best condition of the enzyme trypsin hydrolysis,the volume ratio of the enzyme solution to TCA solution was 4: 1,the shaking time was 10 min,and the settling time was 30 min. Conclusion: The optimal method of deproteination was better,which provided a reference for the further study of the preparation technology of Pinellia Ternate polysaccharides.
