化瘀祛痰方对高脂血症大鼠肝脏PI3K/Akt信号通路影响研究 被引量:6

Effect of Huayu Qutan Decoction on Hepatic PI3K/Akt Signaling Pathway in Hyperlipidemic Rats

摘要 目的:探讨化瘀祛痰方对高脂血症大鼠肝脏PI3K/Akt信号通路的影响。方法:将50只SD大鼠随机分为正常组、模型组、中药低剂量组、中药中剂量组、中药高剂量组,每组10只。采用高脂饲料喂养进行高脂血症模型复制后,中药低、中、高剂量组分别给予含有生药量为0.45、0.9、1.8 g/mL的化瘀祛痰方药煎剂进行治疗,正常组和模型组给予等量生理盐水灌胃。4周后HE染色观察各组大鼠肝脏形态变化;全自动生化分析仪检测各组大鼠血清TC、TG、LDL-C、HDL-C含量变化;WB法检测各组大鼠肝脏PI3K、Akt、p-Akt蛋白表达水平。结果:与正常组相比,模型组大鼠肝脏脂肪变性明显,TC、LDL-C水平显著升高(P〈0.05),HDL-C水平降低(P〈0.05),TG水平未发生显著改变(P〉0.05);PI3K、p-Akt蛋白表达水平上调(P〈0.05);与模型组相比,中药低、中、高剂量组大鼠肝脏脂肪变性呈不同程度减低,TC、LDL-C水平显著降低(P〈0.05),HDL-C水平升高(P〈0.05),TG水平未发生显著改变(P〉0.05);PI3K、p-Akt蛋白表达水平呈不同程度下调(P〈0.05)。结论:化瘀祛痰方可以通过调节PI3K/Akt信号通路相关蛋白的表达,从而改善高脂血症大鼠肝脏脂质损伤。 Objective:To investigate the effect of Huayu Qutan Decoction on hepatic PI3K/Akt signaling pathway in hyperlipidemic rats.Methods:50 SD rats were randomly divided into normal group,model group,low dose group of Chinese medicine,middle dosage group of Chinese medicine and high dose group of Chinese medicine,10 rats in each group.Fed with high-fat diet for hyperlipidemia model replication.Low and medium and high dose groups were given 0.45 g/mL,0.9 g/mL,1.8 g/mL Huayu Qutan Decoction to treat,normal group and model group were given normal saline orally.After 4 weeks,the morphological changes of liver were observed by HE staining in each group;The changes of serum TC,TG,LDL-C and HDL-C in rats were detected by automatic biochemical analyzer;The expression levels of PI3K,Akt and p-Akt in liver of rats were detected by WB.Results:Compared with the normal group,the liver steatosis of the model group was obvious,the levels of TC and LDL-C increased significantly(P0.05),and the level of HDL-C decreased(P0.05),and the level of TG did not change significantly(P0.05);the expression levels of PI3K and p-Akt were up-regulated(P0.05);Compared with the model group,the liver steatosis of rats in the low,medium and high dose groups decreased in different degrees,The levels of TC and LDL-C decreased significantly(P0.05),and the level of HDL-C increased(P0.05),and the level of TG did not change significantly(P0.05),The levels of PI3K and p-Akt protein were down regulated in different degrees(P0.05).Conclusion:Huayu Qutan Decoction can ameliorate the liver lipid damage in hyperlipidemic rats by regulating the expression of related proteins in PI3K/Akt signaling pathway.
