摘要
目的:合成阿德福韦酯(AD)与水杨酸(SA)的盐型并对其合成方法进行优化,为阿德福韦酯水杨酸盐(ADSA)稳定性研究提供基础依据。方法:采用溶剂法和溶剂球磨法合成AD-SA盐,应用差示扫描量热分析(DSC)、热重分析(TG)与X-射线粉末衍射(PXRD)法对其进行表征。结果:成功合成了AD-SA盐,并利用高效液相色谱法(HPLC)对其合成方法进行优化比较,表明球磨法合成的AD-SA化学纯度高于混悬法合成的AD-SA。结论:合成了AD-SA盐并利用HPLC证明合成AD-SA盐球磨法比混悬法更优越,为AD的稳定性及AD-SA盐的研究提供参考。
Objective: To synthesize adefovir dipivoxil( AD) and its salicylate( SA) and to optimize the synthetic conditions as a basis for studying the stability of adefovir dipivoxil salicylatet( AD-SA). Methods: AD-SA was obtained via suspension and solution ball-milling method,and its structure was determined by DSC,TGA,powder and PXRD,respectively. Results: AD-SA was synthesized successfully. Conclusion: The synthesis method was optimized by HPLC,which provided the basic research for the stability of AD and subsequent study of AD-SA.
