异荭草素对鼠伤寒沙门氏菌的抑制作用及机理研究 被引量:3

摘要 研究了异荭草素对鼠伤寒沙门氏菌的抑菌活性,并初步探究了其抑菌机理,为开发高效低毒的抗菌药物提供理论依据。首先,采用滤纸片法测定异荭草素抑菌圈大小和抑菌率来确定异荭草素的抑菌活性;其次,再通过绘制生长曲线、测定细胞膜通透性和细胞内容物渗漏、总核酸来分析异荭草素的抑菌机理。结果表明,异荭草素浓度与对鼠伤寒沙门氏菌的抑菌率呈正相关;其抑菌机理可能是由于异荭草素抑制了细菌对数生长期的分裂速度,并导致细胞膜渗透性增加和细胞内的电解质外漏,影响细菌生长代谢,从而致死菌体细胞。上述结果说明,异荭草素作为天然抗菌剂有较好的开发应用潜力。 This research showed the antibacterial activity of isoorientin on Salmonella typhimurium and its antibacterial mechanism,which provided a theoretical basis for the development of high efficiency and low toxicity antibacterial drugs. The size ofinhibition zone and the rate of inhibition were determined by filter paper method to determine the antibacterial activity of isoorientin.Antibacterial mechanism of isoorientin was analysed by drawing growth curve and determining the cell membrane permeabilityand leakage of intracellular solution (water soluble protein and total nucleic acid) . The results showed that isoorientinconcentration rate was positively correlated with inhibition of S. typhimurium. Antibacterial mechanism of isoorientin might bethe inhibition of bacteria in logarithmic growth phase splitting speed which led to the increase of cell membrane permeabilityand the leakage of electrolyte inside the cell. The growth and metabolism of bacteria were affected and finally caused cell death.The results indicated that isoorientin has the potential of exploitation and application as natural antibacterial agent.
