大鼠脑损伤CCI打击分级模型的建立与评估

Establishment and evaluation of graded brain injury model by CCI in rats

摘要 目的建立大鼠可控性皮层撞击(CCI)模型,并观察不同程度脑损伤指标的变化。方法 48只大鼠随机均分为四组:C组不予以撞击作为对照;A1、A2和A3组均给予速度为4m/s打击,时间150ms,打击深度分别为1、2和3mm。观察各组大鼠体重变化、昏迷程度,采用干湿重法测量脑水肿,Morris水迷宫法检测学习记忆功能变化。结果脑损伤程度越重,大鼠体重下降越明显,昏迷时间和潜伏期越长,脑水肿程度越重,空间探索能力越差(P<0.05)。结论 CCI模型重复性好,能较好地复制分级损伤。 Objective To establish a controlled cortical impact(CCI) rat model and observe the changes of the indexes after different degrees of brain injury. Methods Forty-eight rats were equally randomized into four groups. Group C did not be injured with CCI as the control. The groups of A1, A2 and A3 were implemented CCI with a speed of 4 m/s for 150 ms in the depths of 1 mm,2 rnm and 3 ram, respectively. The changes of weight and degree of coma were observed. The brain edema was measured by wet-dry weight method, and the changes of learning and memory function were tested by Morris water maze. Results With the increase of brain injury, the decrease of weight was more obvious, the time of coma and incubation period were longer, the brain edema was more serious, and the ability of space exploration was worser (P〈0.05). Conclusion CCI animal model has a good replication for duplicating the graded damage of brain injury.
