摘要 [目的]探讨糖尿病模型大鼠胃平滑肌组织纤维化的机制.[方法]取成年SD雄性大鼠,制作糖尿病模型后随机分为正常对照(NC)组和模型4,6,8周组,利用Masson三色染色法检测各组大鼠胃平滑肌组织纤维化情况,利用免疫组织化学染色法检测胃平滑肌组织中波形蛋白的表达水平.[结果]Masson三色染色结果显示,NC组(1.6±0.5)与模型4周组(1.8±0.6)胃平滑肌组织肌间神经丛少见染色;模型6周组(10.1±1.5)与8周组(11.1±1.5)肌间神经丛绿色染色明显,与NC组比较差异均有统计学意义(P<0.01),但两组间差异无统计学意义(P>0.05).免疫组织化学染色结果显示,波形蛋白定位于细胞质内,NC组、模型4,6,8周组的IOD值分别为1.2±0.3,1.4±0.7,2.5±0.9,3.1±1.1,其中模型6,8周组分别与NC组和模型4周组比较差异均有统计学意义(P<0.01).[结论]糖尿病模型大鼠胃平滑肌组织存在肌间神经丛纤维化改变,可能机制认为是波形蛋白参与了该过程. OBJECTIVE To explore the fibrosis mechanism of gastric smooth muscle tissue of rats with diabetes.METHODSAfter the establishment of diabetes model,adult SD male rats were divided into normal control(NC)group,model four weeks(DM4 W)group,model six weeks(DM6 W)group and model eight weeks(DM8 W)group.Masson trichrome staining was used to detect the fibrosis of gastric smooth muscle tissue,and immunohistochemistry was used to detect the expression of vimentin in gastric smooth muscle tissue.RESULTS The results of Masson trichrome staining showed that the staining of myenteric plexus of gastric smooth muscle tissue was rare in the NC(1.6±0.5)and DM4 Wgroup(1.8±0.6),while the staining was obvious in the DM6 Wgroup(10.1±1.5)and DM6 Wgroup(11.1±1.5),compared with the NC group the differences were statistically significant(P〈0.01),but there was no statistically significant difference between the two groups(P〉0.05).The results of immunohistochemical staining showed that vimentin was located in the cytoplasm,IOD value of the NC,DM4W,DM6W and DM8W group was 1.2 ± 0.3,1.4 ± 0.7,2.5 ± 0.9 and 3.1±1.1 respectively,there were statistically significant differences between the DM6W and DM8W group and the NC and DM4W group(P〈0.01).CONCLUSION There existed myenteric plexus fibrosis in gastric smooth muscle tissue of diabetic rat,and the mechanism might be the participation of vimentin in the process.
