摘要
[目的]研究木脂素对胃癌SGC-7901细胞增殖和迁移的影响及其机制.[方法]利用MTT、克隆实验、划痕实验、Transwell小室实验和Western blot法检测木脂素对胃癌SGC-7901细胞增殖、迁移及对Ezrin和P-Ezrin蛋白表达的影响.[结果]MTT检测结果显示,木脂素可抑制胃癌SGC-7901细胞的增殖,且具有浓度和时间依赖性(P<0.05,P<0.01);划痕实验和Transwell小室实验结果显示,木脂素可抑制细胞横向和纵向迁移能力,与对照组比较差异有统计学意义(P<0.05),且具有浓度依赖性;Western blot检测结果显示,木脂素可显著下调Ezrin和P-Ezrin蛋白的表达(P<0.01).[结论]木脂素可抑制胃癌SGC-7901细胞的增殖和迁移,机制认为与下调Ezrin和P-Ezrin蛋白的表达有关系.
OBJECTIVE To study the effects of lignans on proliferation and migration of gastric cancer SGC-7901 cells and its mechanism.METHODS MTT,clone assay,wound healing,Transwell assay and Western blot were used to detect the effects of lignins on proliferation,migration and the expressions of Ezrin and P-Ezrin protein in gastric cancer SGC-7901 cells.RESULTS MTT results showed that the lignins inhibited the proliferation of gastric cancer SGC-7901 cells and had a concentration and time dependence（P〈0.05,P〈0.01）.Wound healing and Transwell assay results showed that the lignins inhibited horizontal and vertical migration of cells and had a concentration dependence,as compared with the control group the difference was statistically significant（P〈0.05）.Western blot results showed that lignans could obviously down-regulate the expressions of Ezrin and P-Ezrin（P〈0.01）.CONCLUSION Lignans could inhibit the proliferation and migration of gastric cancer SGC-7901 cells,the mechanism might be related with the down-regulation of Ezrin and P-Ezrin.
