缺氧环境下利舒康胶囊对大鼠心肌组织的保护作用 被引量:9

Protective Effects of Lishukang Capsule on Rats Myocardial Tissue Under Hypobaric Hypoxia

摘要 目的研究利舒康胶囊对模拟高原减压缺氧大鼠心肌组织的保护作用。方法将60只大鼠随机分为正常对照组、缺氧模型组、红景天胶囊组和低、中、高剂量利舒康胶囊组,采用低压低氧动物实验舱模拟海拔8000m高原环境,减压缺氧12 h,观察缺氧大鼠心肌组织病理显微结构的变化情况,并检测心肌组织中抗氧化应激、能量代谢相关生化指标。结果与正常对照组相比,缺氧后大鼠心肌组织出现明显的病理损伤,心肌组织中丙二醛(malonaldehyde,MDA)、过氧化氢(hydrogen peroxide,H_2O_2)、乳酸(lactic acid,LD)含量及乳酸脱氢酶(lactic dehydrogenase,LDH)活性明显升高(P<0.05或0.01),一氧化氮(nitric oxide,NO)含量虽有所升高,但无统计学差异(P>0.05),超氧化物歧化酶(superoxide dismutase,SOD)、过氧化氢酶(catalase,CAT)、谷胱甘肽过氧化物酶(glutathione peroxidase,GSH-PX)活力均显著性降低(P<0.05或0.01)。给予中、高剂量利舒康胶囊后病理损伤情况明显减轻,心肌组织中MDA、H_2O_2、NO较缺氧模型组均明显下降(P<0.05或0.01),SOD、CAT活性均明显升高(P<0.05),高剂量利舒康胶囊组GSH-PX活力较缺氧组升高明显,LD含量及LDH活性降低明显(P均<0.05)。给予红景天胶囊后病理损伤情况也有所减轻,与缺氧模型组相比,心肌组织中MDA、H_2O_2均明显下降(P<0.01),SOD、CAT、GSH-PX活力均明显升高(P<0.05或0.01),而NO、LD含量及LDH活性则无统计学差异(P>0.05)。结论利舒康胶囊对模拟高原减压缺氧大鼠造成的心脏损伤具有良好的保护作用,与红景天胶囊相当,且在清除自由基和改善机体能量代谢方面更加突出,其机制可能与清除过量自由基,提高机体抗氧化能力,提高乳酸代谢转化,促进无氧酵解,改善机体能量代谢有关。 Objective To investigate the effect of Lishukang capsule on myocardial tissue in hypobaric hypoxia rats.Methods Sixty rats were randomly divided into normal control group,hypoxia model group,Rhodiola capsule group and low,medium,high dosage of Lishukang capsule groups.The rats were exposed to hypoxia for 12 hours at simulating high altitude of 8000 m.The myocardial histopathological changes in hypoxic rats were observed,and the anti-oxidative stress and the related biochemical indicators were detected.Results Compared with normal control group,the myocardial tissues of the hypoxia model group were severely damaged.The malonaldehyde(MDA),hydrogen peroxide(H_2O_2),lactic acid(LD)levels and lactic dehydrogenase(LDH)activity of myocardial tissues were significantly increased(P0.05 or 0.01).The content of nitric oxide(NO)also increased(P0.05).The activities of superoxide dismutase(SOD),catalase(CAT),glutathione peroxidase(GSH-PX)were decreased(P0.05 or 0.01).Pre-treated with medium,high dosage of Lishukang capsule,the pathological injuries were obviously reduced.Compared with hypoxia model group,the MDA,H_2O_2,NO were decreased,the activities of SOD,CAT increased(P0.05 or 0.01),and GSH-PX activity of high dosage of Lishukang capsule group was significantly higher than that of hypoxia model group,LD levels and LDH activity decreased(P0.05).Pre-treated with Rhodiola capsule,the pathological injuries were reduced.Compared with hypoxia model group,the MDA,H_2O_2 levels were decreased,and the activities of SOD,CAT,GSH-PX were increased(P0.05 or 0.01),but there were no significant difference in NO,LD levels and LDH activity(P0.05).Conclusion Lishukang capsule has protective effect on heart injury induced by hypobaric hypoxia in rats.It is equivalent to Rhodiola capsule,and more prominent in eliminating free radicals and improving the energy metabolism.Its mechanism may attribute to scavenge excess free radicals,improve antioxidant capacity,promote anaerobic glycolysis and ameliorate energy metabolism.
