内质网应激在脂蛋白修饰致动脉粥样硬化中作用机制的研究进展 被引量:4

摘要 动脉粥样硬化脂蛋白修饰导致的血管壁内皮细胞下脂质沉积是动脉粥样硬化发生的始动环节。内质网是细胞内多种信号通路的关键调节位点,对蛋白质成熟、正确折叠、分泌和维持钙离子稳态等发挥重要作用。缺血、氧化应激、钙离子稳态失衡、蛋白质错误折叠及正常蛋白质过表达、蓄积等均可导致内质网功能障碍,出现未折叠蛋白反应,引起内质网应激。内质网应激是多种应激过程的共同通路,涉及多个信号通路及相关基因的表达、调控。内质网应激在动脉粥样硬化的发生、发展过程中发挥重要作用,通过氧化修饰、糖基化修饰、糖氧化修饰、氨甲酰化修饰等介导脂蛋白修饰是其主要作用机制,但确切机制尚未完全阐明。
