摘要
目的通过高压静电纺丝法制备PLGA/HA/羊毛角蛋白复合支架并对其理化性能、细胞相容性进行表征。方法先将聚乳酸-羟基乙酸共聚物(PLGA)与羟基磷灰石(Hydroxyapatite,HA)、羊毛角蛋白复合,采用高压静电纺丝法制备PLGA/HA/羊毛角蛋白复合支架,采用扫描电子显微镜(SEM)、红外光谱(FTIR)、万能力学测试机以及细胞培养等分析复合支架的理化性能及细胞相容性。结果 PLGA/HA/羊毛角蛋白复合支架是由长而连续的超细纤维相互交织构成的三维多孔结构,HA、羊毛角蛋白颗粒基本都被包裹于纤维内部;PLGA/HA/羊毛角蛋白复合支架的拉伸强度、断裂伸长率和杨氏模量的平均值分别为4.16MPa、13.96%和135.55MPa;SD大鼠骨髓间充质干细胞在PLGA/MWNTs/羊毛角蛋白复合支架表面生长、增殖良好,HA和羊毛角蛋白的加入有利于细胞的粘附和生长。结论通过高压静电纺丝法制得的PLGA/HA/羊毛角蛋白复合支架具有良好的表面形貌、力学性能和细胞相容性,在组织工程领域有较大的应用潜力,尤其是作为组织工程的支架用于组织再生。
Objective To prepare the PLGA/HA/wool keratin composite scaffold by electrospinningand study its physical and chemical properties and cell biocompatibility.Methods The PLGA/HA/wool keratin composite scaffoldwas prepared by electrospinning;then,SEM,FTIR,universal mechanical testing machine and cell culture were used for characterization.Results The PLGA/HA/wool keratin composite scaffold,composed of long and continuous superfine fibers,had a three-dimensional and porous structure.HA and wool keratin particles were wrapped inside the fibers;SD rat bone marrow mesenchymal stem cells grew well on the surface of PLGA/HA/wool keratin composite scaffold,and the addition of HA and wool keratin was beneficial to cell adhesion and growth.Conclusion The electrospun PLGA/HA/wool keratin composite scaffold has good surface morphology,mechanical properties and biocompatibility,and it may have great potential applications in the field of tissue engineering,especially as a scaffoldfor tissue regeneration.
