摘要
本试验采用反复冻融法、酶裂解法和试剂盒法分别提取猕猴肠道菌群的总DNA,根据提取的DNA浓度和纯度最终确定酶裂解法为较好的方法。将酶裂解法提取的DNA进行ERIC-PCR扩增,结果显示:同年龄组猕猴粪样的ERIC条带数量和位置变化不大;幼年组猕猴的菌群种类较丰富,但优势菌群不明显;青年组猕猴的菌群种类和优势菌群最为丰富;老年组猕猴的菌群种类最少。
The experiment using repeated freeze-thaw method, enzymatic lysis method and reagent box method to extract the total intestinal microflora DNA of macaque, according to the concentration and purity of DNA, we determined enzymatic lysis method was the better way. The DNA extracted by enzymatic lysis method was amplified by ERIC-PCR, the results showed that the number and location of ERIC bands at the same age group changed little, microflora species of childhood macaque were rich, but the dominant microflora was not obvious; microflora species and dominant microflora of youth macaque were richest, mi- croflora species of agedness macaque were least.
