川陈皮素诱导人卵巢癌细胞(HO8910)的凋亡效应

Effects of nobiletin on apoptosis of human ovarian cancer cells(H08910)

目的探讨川陈皮素诱导浆液性上皮性卵巢癌细胞(HO8910)凋亡的作用及分子机制。方法采用MTT检测川陈皮素对HO8910细胞和正常卵巢上皮细胞(IOSE80)生长活力的影响,流式细胞术检测HO8910细胞凋亡率,吖啶橙染色检测HO8910细胞核的形态,JC-1染色判断川陈皮素是否通过线粒体途径诱导HO8910细胞凋亡,Western blot检测Bad、Bcl-xl、Akt、p-Akt、pro-caspase 9和pro-caspase 3蛋白的表达。结果川陈皮素选择性地影响HO8910细胞的活力;当川陈皮素浓度为80μg/mL时诱导的HO8910细胞凋亡率上升;川陈皮素浓度为80μg/mL时作用24 h细胞核发生明显聚集;川陈皮素以浓度依赖方式使细胞线粒体膜电位下降;川陈皮素上调了促凋亡蛋白Bad、pro-caspase 9和pro-caspase 3的表达,下调了抗凋亡蛋白Bcl-xl、Akt和p-Akt的表达。结论川陈皮素可通过线粒体途径,抑制PI3K/Akt信号通路激活,诱导HO8910细胞凋亡。

Objective To study er cancer cells （HO8910）and its the effect of nobiletin on human related molecular mechanisms. serous epithelial ovarian canc- Methods The effect of nobi- letin on the activity of human ovarian cancer cells （HO8910） and normal ovarian epithelial cells （IOSE80） was detected by M3T assay. Flow cytometry was used to analyse the apoptosis rate of HO8910 cells. The morphological changes of the nucleus of HO8910 cells were observed by acridine orange staining. JC-1 staining was used to determine whether nobiletin induced HO8910 cell apoptosis through mitochondrial pathway. The levels of Bad, Bcl-xl, Akt, p-Akt, pro-caspase 9 and pro-caspase 3 were detected by Western blot. Results Nobiletin selectively affect the activity of HO8910 cells; Nobiletin （80 μg/mL） significantly increased apoptosis rate and nucleus aggregation of HO8910 cells at 24 h; Nobiletin decreased mitochondrial mem-brane potential of HO8910 cells in a concentration dependent manner; Nobiletin upregulated the expression of mitochondrial apoptosis-related protein Bad, pro-caspase 9, and pro-caspase 3, and downregulated the expression of antiapoptotic protein Bcl-xl, Akt and p-Akt. Conclu- sion Nobiletin can induce apoptosis in HO9810 cells through the mitochondrial pathway, and PI3K/Akt signaling pathway may be involved in it.
