摘要
目的研究CPEB3对小鼠大脑神经细胞成熟的影响及其潜在机制。方法采用水迷宫实验对CPEB3转基因小鼠及对照组的学习记忆能力进行测试;采用Western blot方法检测CPEB3转基因小鼠和对照小鼠海马神经细胞指标DCX以及成熟神经细胞指标的NeuN表达。结果在CPEB3转基因小鼠的大脑中神经细胞的数目和成熟神经元数目要高于对照组小鼠。CPEB3高表达时只有ApoE4的表达受到抑制,CPEB3的表达降低后ApoE4表达升高,同时Western blot检测到DCX和NeuN都显著下降。结论 CPEB3通过抑制ApoE4的释放促进小鼠大脑神经细胞成熟。
Objective To research underlying mechanism. Methods CPEB3 impact on neuron maturation in mice brain and the CPEB3 transgenic mice and wild type mice were tested by Morris water maze to detect the learning and memorial function, and the expression of neural marker DCX and mature neural marker NenN of CPEB3 transgenic mice and wild type mice in vivo were detected by Western blot. Results The number of neurons and mature neurons in CPEB3 transgenic mice was higher than that in wild type mice. Over-expression of CPEB3 in- hibited the expression of ApoE4. Down-expression of CPEB3 by sh-RNA inhibited the expres- sion of DCX and NeuN, but induced the expression of ApoE4. Conclusion CPEB3 promotes neuron maturation in mice brain via inhibition of ApoE4.
