摘要
目的:探索在不同浓度及作用时间的脂多糖刺激下MT1-MMP在人牙周韧带细胞中的表达。方法:体外培养人牙周韧带细胞,取4~7代细胞分别以脂多糖梯度浓度和梯度时间进行干预,利用Western Blot和实时定量荧光PCR检测细胞中MT1-MMP在蛋白及基因水平上的表达。结果:Western Blot结果示脂多糖刺激下人牙周韧带细胞中MT1-MMP的蛋白表达强度升高,并在一定范围内呈现浓度和时间依赖性(P<0.05);定量PCR结果显示MT1-MMP mRNA的表达变化趋势与Western Blot结果相一致(P<0.05)。结论:脂多糖刺激下人牙周韧带细胞中的MT1-MMP表达显著升高,MT1-MMP可能在牙周组织炎性反应中发挥一定作用。
Objective: To investigate the effects of lipopolysaccharide in graded concentration and stimulating time on MT1-MMP expression in human periodontal ligament cells. Methods: Cultured h PDLCs were stimulated with lipopolysaccharide in concentration and time gradient respectively. Western Blot and quantitative real-time PCR were employed to determine MT1-MMP expressions at protein and gene level. Results: LPS stimulation reduced the expressions of MT1-MMP protein and mRNA,and the regulatory effects of LPS exhibited a certain concentration and time dependence( P 〈 0. 05). Conclusion:MT1-MMP expressions were significantly enhanced in LPS-stimulated h PDLCs,MT1-MMP may be involved in inflammatory reactions of periodontal tissues.
