摘要
目的:检测GSK3β,p-GSK3β(Ser9)及NF-κB p65在人慢性根尖周病变组织中的表达情况。方法:收集人慢性根尖周炎病损组织19例和正常根尖周组织标本6例,分别用于组织学及免疫组织化学染色研究,并提取蛋白进行Western Blot分析,以观察GSK3β,p-GSK3β(Ser9)及NF-κB p65在人根尖周组织中的表达。结果:HE染色显示人慢性根尖周病损组织主要由成纤维细胞、炎症细胞和新生的毛细血管组成,可见大量炎性细胞浸润,主要包括淋巴细胞,中性粒细胞,单核细胞等。免疫组织化学染色结果显示GSK3β,p-GSK3β(Ser9)及NF-κB p65在人慢性根尖周肉芽肿及囊肿中均有表达,并且主要在炎性细胞,血管内皮细胞及成纤维细胞上表达。Western Blot结果发现在正常根尖周组织中仅有少量GSK3β,p-GSK3β(Ser9)及NF-κB p65蛋白表达,且NF-κB p65主要在胞浆蛋白表达;与正常根尖周组织相比,慢性根尖周炎组织中GSK3β,NF-κB p65的蛋白表达显著增多,p-GSK3β(Ser9)表达也略有增多,但幅度不大,与此同时,NF-κB p65的表达发生核转位由胞浆转移到胞核。结论:人慢性根尖周病变组织中有大量GSK3β,p-GSK3β(Ser9)及NF-κB p65阳性细胞的表达,提示其可能参与人慢性根尖周炎病变发展过程。
Objective: The purpose of the present study was to investigate the expression and localization of GSK3β,p-GSK3β( Ser 9) and NF-κB p65 in the human chronic periapical lesions. Methods: Nineteen patients with chronic periapical lesions and 6 cases of normal periapical tissue samples were collected. The histopathological changes of periapical lesions,and immunohistochemical analysis were observed to determine the presence of GSK3β,p-GSK3β( Ser 9) and NF-κB p65.Western Blot analysis were used to examine the protein of GSK3β,p-GSK3β( Ser 9) and NF-κB p65. Results: Composed of fibroblasts,inflammatory cells and new capillaries,It could be observed that a large amount of inflammatory cells infiltrate in the periapical lesions,such as lymphocytes,neutrophils and monocytes. From the immunohistochemical staining,GSK3β,pGSK3β( Ser 9) and NF-κB p65 positive cells expressed in periapical lesions,and they are mainly expressed in inflammatory cells,endothelial cells and fibroblasts. Western Blot analysis revealed that compared with healthy periapical samples,the expressions of GSK3β and NF-κB p65 protein were remarkable elevated in periapical lesions,the expression of p-GSK3β( Ser9) protein also slightly increased. Conclusion: GSK3β,p-GSK3β( Ser 9) and NF-κB p65 can be observed in periapical lesions,it is suggested that they may be involved in the development of chronic periapical periodontitis.
