摘要
目的:探讨维生素B_3(Vit B_3)促中性粒细胞生成的可能机制。方法:取21只健康雌性SD大鼠,随机分为3组(n=7):对照组(灌服0.9%NaCl,2 mL/d,7 d)、rhG-CSF组(皮下注射rhG-CSF,10μg/d,7 d)、Vit B_3组(灌服Vit B_3,500 mg/kg·d^(-1),7 d)。检测干预前及干预后7 d中性粒细胞计数(ANC),应用Bio-Plex悬液芯片系统检测粒细胞-巨噬细胞集落刺激因子(GM-CSF)、肿瘤坏死因子-α(TNF-α)、白介素-1β(IL-1β)、单核细胞趋化蛋白1(MCP-1)、巨噬细胞炎性蛋白-1α(MIP-1α)及肿瘤生长相关因子GRO/KC水平。结果:Vit B_3组和rhG-CSF组ANC较干预前及对照组显著升高(P<0.05),但两组比较差异无统计学意义(P>0.05)。3组干预前血清各细胞因子水平比较,差异无统计学意义(P>0.05);干预7 d后,Vit B_3组TNF-α、IL-1β、MCP-1、GRO/KC明显升高,rhG-CSF组TNF-α显著升高,Vit B_3组IL-1β、MCP-1、GRO/KC明显高于rhG-CSF组,差异均有统计学意义(均P<0.05)。结论:Vit B_3可诱导大鼠中性粒细胞生成,其机制可能与增加炎性因子TNF-α、IL-1β、MCP-1和GRO/KC分泌有关。
Objective: To investigate the effect and mechanism of Vitamin B3 (Vit B3)on granulopoiesis in nor- mal rats. Methods: 21 healthy female SD rats were randomly divided into three groups (n = 7 per group): the control group in which the rats were given 9% NaCl via gavage, the rhG-CSF group in which the rats were giv- en 10 μg/d rhG-CSF via subcutaneous injection for 7 days, and the Vit B3 group in which the rats were given 500 mg/kg rhG-CSF for 7 days. The neutrophil granulocyte counts were analyzed before treatment and the 7th day af- ter administration. The serum cytokines such as granulocyte macrophage colony stimulating factor (GM-CSF), tumor necrosis factor (TNF)-α, interleukin (IL)-1β, monocyte chemoattractant protein-1 (MCP-1), macro- phage inflammatory protein-1αa (MIP-1α)and rumor growth related factors GRO/KC levels were detected by Bio-Plex suspension array system. Results: The neutrophil counts increased significantly in Vit B3 and rhG-CSF groups (P〈0.05), but there was no significant difference between the rhG-CSF group and the Vit B3 group (P〉 0. 05). The levels of TNF-α, IL-1β, MCP-1 and GRO/KC in serum were significantly increased by Vit B3 admin- istration (P〈0. 05). The serum TNF-a level was increased by G-CSF treatment (P〈0. 05). The levels of IL-1β, MCP-1 and GRO/KC were markedly higher in the Vit B3 group than those in the rhG-CSF group (P〈0.05). Conclusion: Vit B3 could result in increasing absolute neutrophil count in healthy rats, and the mechanism might be related to the elevation of serum inflammatory factors TNF-α, IL-1β, MCP-1 and GRO/KC levels.
