溃愈汤对溃疡性结肠炎模型大鼠结肠氧化指标及血清免疫球蛋白表达的影响 被引量:1

Effect of Kuiyu Decoction on Colonic Oxidation Index and Serum Immunoglobulin Expression in UC Model Rats

摘要 为探讨溃愈汤对溃疡性结肠炎(UC)模型大鼠的免疫调节机制及在阻断组织脂质过氧化反应中的作用,本研究将78只SD大鼠分为A(空白对照组)、B(生理盐水组)、C(溃愈汤组)和D(芍药汤组)4组,A组12只,B、C、D组各22只,每组雌雄各半。B、C、D组建立UC模型大鼠,造模成功后分别应用生理盐水、溃愈汤、芍药汤灌肠治疗1个月;同时A组予生理盐水灌肠。对比各组治疗效果、大鼠主要症状改善情况,以及结肠组织丙二醛(MDA)、超氧化物歧化酶(SOD)及血清中分泌型免疫球蛋白A(SIgA)、免疫球蛋白G(IgG)含量的变化。结果显示,与A组相比,B、C、D组大鼠结肠黏膜损伤程度评分、血清sI—gA和IgG含量及结肠组织MDA含量均升高,结肠组织s0D含量降低,P〈0.01。与B组相比,C组和D组大鼠结肠黏膜损伤程度评分、血清SIgA和IgG含量及结肠组织MDA含量均降低,结肠组织SOD含量升高,P〈O.01。C组与D组比较,各指标差异亦有统计学意义,P〈0.01,C组各指标更接近A组。结果表明,溃愈汤灌肠对UC大鼠的治疗效果显著,其治疗作用可能是通过正向干预免疫功能、阻断组织脂质过氧化反应实现的。 In order to explore the effect of Kuiyu decoction on the immunoregulation mechanism and in blocking tissue lipid peroxidation in ulcerative colitis(UC) model rats, 78 SD rats were divided into 4 groups,group A (blank control group),group B(normal saline group),group C (Kuiyu decoction group), group D(Shaoyao decoction group).Group A was 12 rats and the other three groups were 22 rats respec- tively,each group had half male and half femal. Group B, C and D were established UC model rats; after successful establishing rats model, the three groups all accepted normal saline, Kuiyu decoction, Shaoyao decoction for enema for 1 month respectively;at the same time group A adopted normal saline for enema. The efficacy, the improvement status of the main symptoms of rats, and the content changes of the malond- ialdehyde(MDA),the superoxide dismutase (SOD) in colonic tissue and serum SIgA and IgG were com- pared among the three groups.As results,the score on rat's colonic mucosa injury degree,the levels of SIgA and IgG in serum and the content of MDA in colonic tissue increased, and the content of SOD in colonic tissue decreased in group B,C and D compared with group A ( P〈0.01). Compared with group B,the score on rats'colonic mucosa injury degree,the levels of SIgA and IgG in serum and the content of MDA in co- lonic tissue were decreased in group C and group D,and the content of SOD in colonic tissue increased( P〈0.01). Compared between group C and group D, there were significant differences in all indexes( P 〈0.01). The indexes of group C were closer to group A. The results show that the efficacy of Kuiyu decoction enema for the treatment of UC is significant, and its therapeutic effect could be achieved by positively intervening the immune function and blocking tissue's lipid peroxidation.
