香薷散对热应激蛋鸡肠道菌群的影响 被引量:4

Effect of elsholtziaeon powder on intestinal flora of egg-laying hens under heat stress

摘要 为探讨香薷散对热应激蛋鸡肠道菌群的影响,选取30只罗曼蛋鸡,随机平均分为3组,香薷散组日粮中添加1%香薷散,热应激组、空白组饲喂基础日粮。于试验的第1、3、7、14、28天时,提取蛋鸡粪便的DNA,进行高通量测序,检测肠道菌群。结果显示,试验的第1、3天时,香薷散能抑制热应激,显著增加OTU的数目(P〈0.05);试验的第1天时,热应激组的Chao1指数、Shannon值显著降低(P〈0.05);PCA、PCo A分析显示香薷散组、空白组聚拢趋势显著,物种组成相似,同时与热应激组有明显差异;热应激可以显著降低试验第1、14天时乳酸杆菌属的丰度比例,第3、28天时Faecalibacterium的丰度比例(P〈0.05);香薷散组第3天时乳酸杆菌属的丰度比例高于热应激组(P〉0.05),第3、7天时Faecalibacterium的丰度比例,第28天时普雷沃菌属的丰度比例显著高于热应激组(P〈0.05);中药香薷散可以增加第1、3、7、28天时拟杆菌属的丰度比例(P〉0.05)。结果表明,香薷散可以改变热应激蛋鸡肠道菌群的多样性和丰富度,调节肠道菌群丰度比例的变化,降低热应激给蛋鸡带来的损伤。 The experiment was investigated the influence of elsholtziaeon powder on intestinal microflora of egg-laying hens under heat stress,30 Roman hens were randomly divided into three groups,Chinese medicine group added 1% Elsholtzia diet powder,heat stress group and the control group were fed basal diet.On the day of 1,3,7,14,and 28 after heat stress,DNA of all fecal samples was extracted and sequenced for high throughput sequencing and detect intestinal flora,respectively.The results showed that on the 1 st and 3 rd day after heat stress,elsholtziaeon powder could inhibit the heat stress and significantly increase the number of operational taxonomic units(OTU) in intestinal flora(P〈0.05).On the 1 st day after heat stress,the result of Chao1 index,Shannon index of heat stress group was significantly decreased(P〈0.05).The PCA and PCo A analysis showed that the Chinese medicine group and blank group gathered trend obviously.The species composition of elsholtziaeon powder group and blank group was similar,both had the obvious difference with the heat stress.Heat stress could significantly reduce the abundance ratio of the 1 st day and 14 th day lactic acid bacteria rod, abundance the ratio of Faecalibacterium on the 3 rd and 28 th day after heat stress(P〈0.05).On the 3 rd day after heat stress,theabundance ratio of elsholtziaeon powder group lactic acid bacteria was increased,the abundance ratio of Faecalibacterium on the 3 rd,7 th day and the abundance ratio of Prevotella spp. on the 28 th day were significantly higher than that of heat stress group(P〈0.05).Meanwhile,elsholtziaeon powder could increase the abundance ratio of Bacteroides on the 1 st,3 rd,7 th,and 28 th day(P〈0.05).These data demonstrated that elsholtziaeon powder could change the diversity and richness of intestinal flora of egg-laying hens under the heat stress,regulate the proportion of intestinal flora abundance,and reduce the damage caused by heat stress to egg-laying hens.
