摘要
目的研究金匮肾气丸对庆大霉素损伤下豚鼠听力的保护作用和耳蜗螺旋神经节元A型γ-氨基丁酸受体(gamma-aminobutyric acid receptor,GABA_AR)表达的影响。方法将30只(60耳)双侧耳廓反射正常的DHP白毛红目雌性豚鼠随机平分3组:正常对照组(control group,CG)、模型组(gentamicin model group,GM)和治疗组(treatment group,TG),造模12天后,行畸变产物耳声发射(DPOAE)与听性脑干反应(ABR)测试,并取双侧耳蜗行免疫印迹法(western blot)检测各组豚鼠耳蜗GABA_ARα1的表达情况。结果 GM组和TG组的ABR阈值均极显著高于CG组(P<0.01),且GM组的ABR阈值显著高于TG组(P<0.05)。GM组和TG组的DPOAE幅值在不同频率均极显著低于CG组(P<0.01)。GM组的耳蜗螺旋神经节GABA_ARα1的表达量极显著低于CG组(P<0.01);TG组的GABA_ARα1的表达量显著高于GM组(P<0.05),但较CG组低(P>0.05)。结论本实验用金匮肾气丸汤剂可通过上调拮抗庆大霉素引起的耳蜗GABA_ARα1蛋白对庆大霉素引起的听力下降起一定的保护作用。
Objective To study the protective effects of Jinguishenqi Pills on the hearing of guinea pigs induced by gentamicin and the expression of gamma-aminobutyric acid receptor(GABAAR) in the cochlear spiral ganglion cells. Methods In this study, 30(60 ears)normal DHP white female red guinea pigs with normal ears were randomly divided into three groups the control group (CG),the model group (GM) and the treatment group(TG). 12 days after modeling,the DPOAE and ABR,and the changes of GABAARα1 expression in cochlear hair cells of guinea pigs were detected. Results The ABR thresholds of GM group and TG group were significantly higher than CG group(P〈0.01), and those of GM group were higher than TG group (P〈0.05).GM group and TG group DPOAE amplitudes at different frequencies were significantly lower than group CG (P〈 0.001). The expression of GABAARα1 in cochlear spiral ganglion alpha 1 in GM group was lower than that in the CG group(P〈0.01); TG group was higher than GM group (P〈0.05) , but it was lower than CG group(P〉0.05).Conclusion The experimental use of JinguiShenqi has certain protective effects by upregulation of antagonistic gentamic in GABAARα 1 protein on gentamicin induced hearing loss.
