摘要
观察黄芪、丹参配伍提取物对心肌梗死大鼠血管内皮生长因子(vascular endothelial growth factor,VEGF)及其受体KDR(kinase insert domain receptor)的调控作用,分析其对心肌梗死大鼠的保护机制。对SD雄性大鼠采用冠状动脉左前降支结扎法复制心肌梗死大鼠模型,随机分为模型组、黄芪、丹参配伍提取物高、中、低剂量组(40、20、10 mg·kg-1·d-1);另设假手术组,各组均为8只。黄芪、丹参配伍提取物各组采用上述对应剂量灌胃给药,模型组和假手术组采用等量生理盐水灌胃,连续饲养28 d后采用ELISA法、RT-PCR法和Western blot法检测分析VEGF及其受体KDR的表达变化。黄芪、丹参配伍提取物各剂量组与模型组相比VEGF及其受体KDR随着剂量加大表达明显升高(P<0.01,P<0.05)。说明:黄芪、丹参配伍提取物可以有效上调VEGF及其受体KDR的表达,从而促进心肌梗死大鼠心肌组织中血管新生作用。
To observe the regulatory effects of Astragalus and Salvia miltiorrhiza extract on VEGF and its receptor KDR in rats with myocardial infarction,and to investigate the protective mechanism of Astragalus membranaceus and Salvia miltiorrhiza on myocardial infarction in rats,male SD rats by left anterior descending coronary artery in rats with myocardial infarction model copy ligation method,were randomly divided into model group,Astragalus and Salvia miltiorrhiza extract with high,medium and low dose group( 40,20,10 mg·kg-1·d-1),sham operation group,each group had 8 rats. Astragalus and Salvia miltiorrhiza Extract the corresponding dose groups with intragastric administration,the model group and sham operation group with normal saline,the expression change of ELISA method,RT-PCR method and Western blot method for detecting VEGF and its receptor KDR by continuous feeding after 28 d. Compared with the model group,the dosage of VEGF and its receptor KDR increased significantly with the increase of dosage( P < 0. 01,P < 0. 05). The combination of Astragalus and Salvia miltiorrhiza extract can effectively up regulate the expression of VEGF and its receptor KDR,thereby promoting angiogenesis in myocardial tissue of rats with myocardial infarction.
