微型月季花粉离体萌发培养基组分的优化 被引量:2

摘要 采用液体培养法研究了不同培养基组分对微型月季Unconditional Love花粉萌发和花粉管生长的影响,旨在为微型月季花粉活力检测和杂交育种提供参考。结果表明,在液体培养基内添加蔗糖、硼酸、硝酸钙、硫酸锌、硫酸镁和硝酸钾,均能促进微型月季Unconditional Love花粉萌发和花粉管生长;在一定范围内,花粉萌发率和花粉管生长长度均随培养基组分添加量的增加而增加,但超过一定添加量时,则出现抑制作用;在此基础上通过正交试验,比较了蔗糖、硼酸和硝酸钙对花粉萌发和花粉管生长的影响,结果表明,最适合微型月季花粉萌发的蔗糖、硼酸和硝酸钙3种组分的培养基配方为蔗糖150 g/L+H_3BO_3125 mg/L+Ca(NO_3)_2·4H_2O 100 mg/L。 The effects of different medium components on pollen germination and pollen tube growth of Rosa hybrida var.Minina Unconditional Love were studied wih liquid culture,so as to provide reference for pollen viability detection and hybridization breeding of Rosa hybrida var.Minina.The results showed that in the liquid medium with sucrose,H_3BO_3,Ca(NO_3)_2·4 H_2O,ZnSO_4·7 H2_O,MgSO_4·7 H_2O,KNO_3 could promote the Rosa hybrida var.Minina Unconditional Love pollen germination and pollen tube growth in a certain range,but this function was inhibited if exceeded a certain amount.Orthogonal test based on the effects of sucrose,H_3BO_3,and Ca(NO_3)_2·4 H_2O on pollen germination and pollen tube growth were compared and screened out the most suitable culture medium for pollen germination of Rosa hybrida var.Minina Unconditional Love,which was sucrose 150 g/L+H_3BO_3125 mg/L+Ca(NO_3)_2·4 H_2O 100 mg/L.
