摘要 本研究从黑麦中克隆了一个热激蛋白Hsp90基因,暂命名为ScHsp90-1,并对其序列进行分析。结果显示,该基因cDNA序列全长2 103 bp,共计编码700个氨基酸,蛋白质分子量约80.47 k D。序列同源性分析表明,ScHsp90-1与小麦、玉米、水稻等物种的热激蛋白同源性较高;进化树分析表明ScHsp90-1与小麦的Ta Hsp90亲缘关系最近。利用实时荧光定量PCR技术对其表达特性分析表明,ScHsp90-1对高温、低温、高盐、干旱等非生物胁迫均有响应,其可能是黑麦的一个胁迫相关基因。 In this study,a Hsp90 gene was isolated from rye,named Sc Hsp90-1. The full c DNA sequence of Sc Hsp90-1 is 2 103 bp,which encodes 700 amino acids with a predicted molecular mass of 80. 47 k D. The sequence analysis showed that Sc Hsp90-1 had higher homology with that of wheat,maize and rice.The phylogenetic analysis indicated that Sc Hsp90-1 was especially similar to Ta Hsp90 from wheat. The real-time quantitative PCR analysis showed that Sc Hsp90-1 had response to high-temperature,cold,high-salt and drought treatments in rye. These results indicated that Sc Hsp90-1 gene might be a stress-related gene of rye.
