摘要
为研究接骨木花粉活力、适宜培养基及花粉贮藏最佳条件。本研究以接骨木花粉为材料,采用3种方法进行花粉活力测定,用5个不同硼酸浓度花粉培养基进行花粉萌发培养,在4个不同温度条件下对接骨木花粉进行贮藏。结果表明:(1)I-IK染色法和TTC氧化还原法测定结果均大于花粉萌发法,两者均不适合测定接骨木花粉的萌发。(2)接骨木花粉的最佳培养基为15%蔗糖+0.01%硼酸+1%琼脂。(3)接骨木花粉的最佳贮藏温度为-50℃。
To study the viability,suitable medium and optimal storage conditions of pollen of Sambucus williamsii Hance,3 methods were conducted to determine the pollen viability,5 mediums with different concentrations of boric acid were used for pollen germination,and the S. williamsii pollen was stored in 4 different temperature conditions. The results showed that:( 1) the viability determination results of TTC oxidation reduction method and I-IK staining method were greater than that of germination method,and they both were not suitable for determining the pollen germination vigor;( 2) the best medium for S. williamsii pollen was15% sucrose + 0. 01% boric acid + 1% agar;( 3) the best storage temperature for S. williamsii pollen was -50℃.
