利湿通痹汤及其拆方对高尿酸血症合并急性痛风性关节炎大鼠疗效机制研究

Action Mechanism Study of Lishi Tongbi Decotion and Its Seperated Groups on Hyperuricemia Rats Complicated by Acute Gouty Arthritis

目的:观察利湿通痹汤及其拆方对高尿酸血症合并急性痛风性关节炎(acute gouty arthritis,GA)大鼠的疗效并探索其作用机制。方法:42只大鼠分为利湿通痹汤全方组,祛风通经组,清热利湿组,活血化瘀组,西药组,模型组和对照组。先建立大鼠高尿酸血症模型,然后在后关节腔内注射尿酸钠诱导大鼠GA模型,对照组予以等量生理盐水。以缚线法测量大鼠踝关节同一部位周径(取均值),观察其关节肿胀度;于造模48 h后腹腔静脉采血,测定血清尿酸(UA)、白介素-1β(IL-1β)水平及黄嘌呤氧化酶(XOD)活力。结果:利湿通痹汤及其拆方均可不同程度降低高尿酸血症合并GA大鼠关节肿胀度;降低血清IL-1β、UA水平,抑制XOD活力,以全方组最为明显(P<0.01)。结论:利湿通痹汤及其拆方可通过降低IL-1β与UA水平、抑制XOD活力而发挥较好的治疗作用,拆方中以清热利湿类药物作用较强。

Objective: To observe the effect of Lishi Tongbi decoction and its separated groups on hyperuricemia rats complicated by acute gouty arthritis( GA),and to explore its action mechanism. Methods: 42 rats were randomly divided into seven groups,i. e. the blank control group( group A),the model group( group B),the group of whole original formula( group C),the group of expelling wind and dredging meridian( group D),the group of clearing heat and dampness( group E),the group of activating blood and removing stasis( group F). Models of hyperuricemia were firstly established,and then the rats were induced into GA model with intra-articular injection of sodium urate. The blank control group was injected with equal dose of normal saline. The mean value of ankle perimeter was measured to observe the joint swelling degree. Blood sample was collected after 48 h of modeling to measure the serum levels of uric acid( UA) and interleukin 1-β( IL-1β),as well as activities of xanthine oxidase( XOD). Results: Lishi Tongbi decoction could reduce the swelling of joints in rats with hyperuriccemia complicated by acute GA to varing degrees,it could reduce the serum levels of IL-1β and UA,and inhibit the activity of XOD; the effect of group C was most obvious( P < 0. 01).Conclusion: Lishi Tongbi decoction and its separated groups can play a good therapeutic effect on hyperuricemia complicated by GA by decreasing the levels of IL-1β and UA,and inhibiting the activity of XOD; group E has the strongest effect among the separated groups.
