摘要 目的采用响应曲面法优化超声提取诸葛菜总黄酮工艺及研究其抗氧化活性。方法在单因素试验基础上,以料液比、提取时间、超声温度、乙醇浓度为自变量,总黄酮提取率为响应指标,采用响应曲面法研究各自变量及其交互作用对总黄酮提取率的影响。通过测定诸葛菜总黄酮对DPPH自由基清除率来考察其抗氧化生物活性。结果优化超声提取诸葛菜总黄酮的工艺为:料液比1∶50,超声时间51min,超声温度38℃,乙醇浓度70%,得到诸葛菜总黄酮平均提取率为21.76mg/g。抗氧化试验结果表明,供试品浓度在10~50μg/ml时,对DPPH自由基清除能力均随着样品溶液浓度增大而增加。结论响应曲面法优化超声提取诸葛菜总黄酮的工艺具有可行性。诸葛菜总黄酮有一定的抗氧化活性。 Objective To optimize the conditions of the ultrasound assisted extraction for the extraction of total flavonoids from Orychophragmus violaceus by response surface methodology,and study on the antioxidant activity. Methods On the basis of single factor experiment,the liquid ratio,extraction time,temperature,concentration of ethanol was used as independent variables. The extraction yield of total flavonoids as index parameter,the variables and their interactions on extraction yield of total flavonoids were studied by response surface methodology. The antioxidant bioactivity was investigated by the scavenging free radical test on DPPH of the total flavonoids. Results The optimal conditions obtained were as follows: ratio of material to liquid 1∶ 50,ultrasonic extraction time 51 min,extraction temperature 38 ℃,ethanol concentration 70%,and the yield of total flavonoids from Orychophragmus violaceus was 21. 76 mg/g. The results of antioxidant tests showed that the scavenging capacity on DPPH was increased in dose-dependent with the concentration of the sample solution at 10 ~ 50 μg/ml. Conclusion The method of response surface methodology to optimize ultrasonic extraction of total flavonoids is feasible. The flavonoids of Orychophragmus violaceus have certain antioxidant activity.
