独正杆接骨膏外用对骨折愈合过程中SMAD1/5、SMURF1mRNA表达的影响

Effect of Duzhenggan Jiegu Ointment on Expression of SMAD1/5 and SMURF1mRNA in Process of Fracture Healing

目的：探讨独正杆接骨膏外用对家兔骨折愈合过程中SMAD1/5、SMURF1mRNA表达的影响,分析其促进骨折愈合的作用机理。方法：建立30只家兔桡骨骨折模型,随机分为实验组和对照组,每组15只。实验组在其患处外敷接骨膏并用小夹板固定;对照组不使用药物,仅常规消毒后进行小夹板固定。分别于给药后第3、6、10周每组随机抽取5只,行X线拍片后处死家兔取患处组织标本。采用实时荧光定量PCR方法测定家兔骨组织中SMAD1/5、SMURF1mRNA的表达,对所得数据进行统计学分析。结果：实验组家兔第3周患处骨组织中SMAD1/5 mRNA表达高于对照组;SMURF1 mRNA表达低于对照组,差异有统计学意义（P〈0.05）。第6周实验组家兔患处骨组织中SMAD1、SMURF1 mRNA的表达与对照组相比无统计学意义（P〉0.05）;SMAD5 mRNA的表达低于对照组,差异具有统计学意义（P〈0.05）。第10周实验组家兔骨组织中SMAD1/5 mRNA的表达显著高于对照组,差异有统计学意义（P〈0.05）;SMURF1 mRNA的表达与对照组相比,无统计学意义（P〉0.05）。结论：独正杆接骨膏外用可在骨折愈合早期促进骨组织中SMAD1/5mRNA的表达,抑制SMURF1mRNA的表达,提高骨折晚期SMAD1/5mRNA的表达量,从而促进骨折愈合,缩短骨折愈合时间。

Objective： To investigate the influence of Duzhanggan Jiegu Ointment on rabbit fracture healing process SMAD1/5 and SMURF1 mRNA expressions and analyze the promoting fracture healing mechanism. Methods： Thirty rabbits were randomly divided into experimental group and control group,with 15 rabbits in each group. In experimental group,we used external application of Jiegu Ointment and fixed with small splints. The control group was without the use of drugs and only used small splint fixing after routine disinfection. Respectively,after administration of the 3 rd,6 th,10 th week,5 were randomly selected in each group. After x-ray photograph,rabbits were killed after the affected area from the tumor specimen. The expressions of SMAD1/5 and SMURF1 mRNA in bone tissue of rabbits were measured by real-time fluorescence quantitative PCR method,and the data was statistically analyzed. Results： In the third week,the expression of SMAD1/5 mRNA in the experimental group was higher than that in the control group and SMURF1 mRNA expression was lower than that of the control group. The difference was statistically significant（ P 0. 05）. At the sixth week,compared with the control group,the expressions of SMAD1 and SMURF1 mRNA in the experimental group were similar to those in the experimental group and there was no statistical significance（ P 0. 05）. SMAD5 mRNA expression was lower than that of the control group and the difference was statistically significant（ P 0. 05）. At the tenth weeks,the expression of mRNA SMAD1/5 in the bone tissue of rabbits was significantly higher than that in the control group,and the difference was statistically significant（ P 0. 05）. Compared with the control group, the expression of SMURF1 mRNA was not statistically significant（ P 0. 05）. Conclusion： Duzhanggan Jiegu Ointment can promote the expression of SMAD1/5 mRNA in the early stage of fracture healing in bone tissue,inhibit the expression of SMURF1 mRNA and improve the expression of SMAD1/5 mRNA in the late fracture so as to promote the fracture healing and shorten the healing time.
