人参皂苷对大鼠成骨细胞增殖及成骨细胞中PINP、β-CTX分泌的影响 被引量:5

摘要 目的:探讨人参皂苷对大鼠成骨细胞增殖及成骨细胞中I型前胶原氨基端前肽(PINP)、β-胶原降解产物(β-CTX)分泌的影响。方法:将出生24 h的SD大鼠头盖骨成骨细胞行体外原代及传代培养,分别在培养液中加入不同浓度(0、1、5、10、20μmol/L)的人参皂苷传代培养21 d,行茜素红染色并于倒置相差显微镜中观察成骨细胞矿化结节,应用Western blotting法测定大鼠成骨组织中PINP、β-CTX表达水平。结果:人参皂苷可促进大鼠成骨细胞增殖及矿化结节形成,且呈时间及浓度依赖性。随着人参皂苷浓度增加及作用时间延长,大鼠成骨细胞中PINP、β-CTX表达水平显著升高(P〈0.05)。结论:人参皂苷可促进大鼠成骨细胞增殖及矿化结节形成,其可能作用机制与人参皂苷可诱导成骨细胞组织中PINP、β-CTX分泌有关。 Objective: To investigate the effects of ginsenoside on the proliferation of rat osteoblasts and the secretion of type I procollagen amino terminal propeptide( PINP) and β-collagen degradation product( β-CTX) in osteoblasts.Methods: Sprague-Dawley( SD) osteoblasts were cultured in vitro and subcultured in vitro for 24 h. Cultured cells were treated with different concentrations( 0,1,5,10,20 μmol/L) for 21 days. The mineralized nodules of osteoblasts were observed under inverted phase contrast microscope. Alkaline phosphatase( PINP) and β-CTX( 10 μmol/L) were detected by Western blotting. Expression. Results: Ginsenoside could promote osteoblast proliferation and mineralized nodules formation in a dose-dependent and time-dependent manner. With the increase of ginsenoside concentration and the prolongation of the time,the expressions of PINP and β-CTX in osteoblasts of rats were significantly increased( P 0. 05). Conclusion: Ginsenoside can promote osteoblast proliferation and mineralized nodules formation in rats. The mechanism may be related to the secretion of PINP and β-CTX in ginsenosides-induced osteoblasts.
