谷胱甘肽过氧化物酶活性和总抗氧化活性在健康人中参考区间的确立

Establishment of glutathione peroxidase activity and total antioxidant status reference intervals in healthy people

目的建立厦门地区健康人中谷胱甘肽过氧化物酶活性(GPX)和总抗氧化活性(TAOS)的参考区间。方法共收集2016年3~8月间厦门市第二医院体检健康的人群253例,年龄13~63岁,其中男性136例,女性117例。样本血液用肝素锂抗凝,在全自动生化分析仪中检测,用酶偶联速率法测定全血中GPX活性,比色法检测血浆中TAOS水平。根据年龄或性别进行分组,应用SPSS19.0软件对数据进行统计分析。结果 GPX项目的批间重复性CV%=3.1%,253例中D最大=0.571,R=8.738,D最大/R<1/3,未发现离群值,P-P图示结果基本呈正态分布,按性别统计:男性(9.044±1.817)k U/L,女性(8.740±1.698)k U/L,Z值=1.374,小于Z判断限值Z*=2.1213,不考虑分组;按年龄统计:13~40岁(8.864±1.751)k U/L,41~63岁(8.962±1.796)k U/L,Z值=0.430,小于Z*,亦不考虑分组。TAOS项目的批间重复性CV%=3.7%,253例中D最大=0.13,R=0.99,D最大/R<1/3,未发现离群值,P-P图示结果基本呈正态分布,按性别统计:男性(1.595±0.161)mmol/L,女性(1.467±0.152)mmol/L,Z值=6.498,大于Z*=2.1213,需要分组确立参考区间;按年龄统计:13~40岁(1.540±0.161)mmol/L,41~63岁(1.530±0.180)mmol/L,Z值=0.452,小于Z*,不考虑分组。结论采用全自动分析仪检测全血GPX和血浆中TAOS的方法完全满足分析要求,经统计确立厦门地区健康人全血GPX的参考区间为5.371~12.435 k U/L;血浆中TAOS的参考区间为男性(1.27~1.92)mmol/L、女性(1.16~1.77)mmol/L。

Objective To establish the reference intervals of glutathione peroxidase （GPX） activity in wholeblood and total antioxidant status （TAOS） in plasma in healthy subjects. Methods A total of 253 healthy subjects whoreceived physical examination in Affiliated No.2 hospital of Xiamen Medical College from March 2016 to August 2016were collected, aged 13~63 years old, including 136 males and 117 females. The samples were treated with heparin lithi-um anticoagulant and detected by automatic biochemical analyzer. The activity of GPX in whole blood was measured byenzyme coupling rate method. The level of TAOS in plasma was measured by colorimetric method. SPSS19.0 was em-ployed for statistical analysis in this study. Results The inter-batch CV% of GPX was 3.1%, with Dmax=0.571, R=8.738, Dmax/R〈1/3. P-P results showed normal distribution： statistics by gender： the male was （9.044±1.817） kU/L andthe female was （8.740±1.698） kU/L, Z=1.374 （less than the limit of Z＊=2.1 213）, not supporting grouping by gender; sta-tistics by age： 13~40 years old was （8.864±1.751） kU/L, 41-63 years old was （8.962±1.796） kU/L, Z=0.430Z＊, indicating reference intervals should be established by gender; statistics by age： 13~40 yearsold was （1.540±0.161） mmol/L, 41~63 years old was （1.530±0.180） mmol/L, Z=0.452 〈Z＊, not supporting grouping by age.Conclusion The procedure for detecting GPX in whole blood and TAOS in plasma is fully dependable. The reference in- tervals of blood GPX activity were 5.371-12.435 kU/L, and the reference intervals of plasma TAOS level were 1.27-1.92 mmol/L in the male and 1.16-1.77 mmol/L in the female.
