摘要
目的 :分析猪囊尾蚴 DNA疫苗 pc DNA3- c C1肌肉接种后在猪体内的组织分布。 方法 :分别于疫苗接种后 1d、7d、4周、8周 ,提取猪骨髓、肾、肝、脑、性腺、肠系膜淋巴结、脾、胸腺、外周血及肌注部位的肌肉和皮肤的总 DNA,通过 PCR的方法检测疫苗质粒在各组织中的分布及随时间变化的情况。 结果 :疫苗接种后的第 1天几乎所有组织中都检测到质粒 ;但随着时间的延续 ,仅在注射部位的肌肉和皮肤中检测到质粒的存在 ,并保持到 8周以上。 结论 :肌注疫苗质粒后 。
Objective:To examine the tissue distribution in porcine following intramuscular administration of the DNA vaccine pcDNA3 cC1. Methods:Samples including bone marrow, kidney, liver, brain, gonad,mesenteric lymph node, spleen, thymus, peripheral blood, skin and muscle of the injection site were obtained 1 d, 7 d, 4 weeks and 8 weeks after intramuscular injection. PCR technique was used to detect the distribution and kinetics of plasmid vaccine in the tissues. Results:After intramuscular administration, plasmid DNA was detected 1 d after in almost all of the tissues, but at later time points it were found only in the muscle and skin at the site of injection, where it persisted for up to 8 weeks. Conclusion:It suggests that plasmid DNA is cleared from peripheral systemic locations in short time following intramuscular injection under the experimental condition.
