巢湖铜绿微囊藻群体mic16s RNA基因季节演变的研究

摘要 为了更深入地了解铜绿微囊藻水华演替机制及其优势维持机理,采用96孔细胞培养板并结合毛细管分离法和极限稀释法分离和纯化巢湖不同铜绿微囊藻藻株,并利用序列(mic16s RNA)设计引物对209F和409R,建立邻接进化树。BLAST结果显示正义链与反义链完全配对的核苷酸长度为250bp左右,差别不大。Clustal W测定结果指出同一季节铜绿微囊藻藻株扩增产物的测序结果有较大一致性,但不同季节只有少部分铜绿微囊藻藻株的序列存在着相似性。 To study the succession mechanism of Microcystis bloom and its dominant maintenance, the 96 well microplates combining the straw separation and utmost dilution culture methods were used. Microcystis aeruginosa strains from different seasons were isolated and purified from Chaohu Lake. The primer pair, 409R and 209F, was designed/or constructing the neighbor-joining tree based on mic-16s RNA. The results analyzed by the BLAST showed that 250 bp paired chains. Clustal W analysis indicated that no significant difference was found between the the PCR amplification fragments from the same season were almost identical, but only a few strains from different seasons showed consistency.
