党参多糖对肾阴虚大鼠抗氧化活性和免疫调节影响 被引量:23

Study on Antioxidant and Immune Regulation Effects of Codonopsis Polysaccharides on Deficiency of Kidney-yin Type Rats

摘要 目的:观察党参多糖的抗氧化活性和对肾阴虚大鼠的免疫调节作用。方法:采用灌胃氢化可的松的方法建立大鼠肾阴虚模型,将成模大鼠随机分为正常对照组、模型组与党参多糖高、中、低剂量组,每组10只。党参多糖高、中、低剂量组分别给予0.5、1、2g/kg党参多糖灌胃,模型组与正常对照组用等体积生理盐水灌胃。持续用药3周后,对5组大鼠的丙二醛(MDA)、超氧化物歧化酶(SOD)、皮质醇(CORT)三种抗氧化指标及免疫球蛋白-G(Ig-G)、免疫球蛋白-M(Ig-M)、白细胞介素-6(IL-6)、白细胞介素-2(IL-2)四种免疫指标进行检测并比较分析。结果:与正常组比较结果显示,模型组大鼠体重,血清中SOD及IL-2的浓度显著降低,而血清中CORT、MDA、IL-6、Ig-G、Ig-M的浓度显著增高(P<0.01);与模型组大鼠比较结果显示,用药组体重有显著增长(P<0.05),但与空白组比较无显著差异(P>0.05),用药组大鼠体内的CORT浓度显著降低,且高剂量组优于低剂量组,组间比较有显著性差异(P<0.05),高剂量用药组大鼠血清中的SOD活性显著升高,而MDA活性则有所降低(P<0.01),而低剂量组有改善趋势,但差异无统计学意义(P>0.05),高剂量组大鼠血清中的IL-2水平升高且IL-6、Ig-G、Ig-M浓度有显著降低(P<0.01)。结论:党参多糖可改善肾阴虚大鼠的免疫状态并可提高其抗氧化能力,且与用药剂量相关。 Objective: To observe the antioxidant activity of Codonopsis pilosula polysaccharides and immunosuppressive effects on rats with kidney-yin deficiency. Methods: The model of kidney-yin deficiency in rats was established by gavage of hydrocortisone. The model rats were randomly divided into normal control group, model group and Codonopsis high, medium and low dose groups, 10 in each group. Codonopsis polysaccharide high, medium and low dose groups were given 0.5,1,2 g/kg ginseng polysaccharides gavage, the model group and normal control group with equal volume of normal saline. Three weeks after the continuous administration, the three antioxidant indexes such as MDA, SOD, CORT and the immunoglobulin-G(Ig-G) Immunoglobulin-M, IL-6 and IL-2 were detected and compared.Results: Compared with the normal group, the body weight, the level of serum SOD and IL-2 in the model group were significantly decreased, while the concentrations of CORT, MDA, IL-6, Ig-G and Ig-M in the model group were significantly increased(P0.01). Compared with the model group rats, the body weight of the treated group increased significantly(P0.05), but there was no significant difference compared with the blank group(P0.05). The concentration of CORT in the treatment group was significantly lower than that in the low-dose group, and there was significant difference between the two groups(P0.05). The activity of SOD in the high dose group was significantly increased, while the MDA activity was decreased(P0.01), while the low dose group was improved, but the difference was not statistically significant(P0.05). Conclusion: Codonopsis polysaccharide can improve the immune status of rats with kidneyyin deficiency and improve its anti-oxidation ability, and with the dosage. The level of IL-2 in the serum of high-dose group increased and the concentrations of IL-6, Ig-G and Ig-M were significantly decreased(P0.01).
