树突状细胞在睾丸原发性淋巴瘤组织中的免疫表型分析

摘要 目的研究树突状细胞(DCs)在睾丸原发性淋巴瘤(PLT)患者睾丸组织中的免疫表型及其分子机制。方法采用免疫组织化学染色法(IHC)检测10例正常人及7例睾丸原发性淋巴瘤患者睾丸组织中CD11c、DC-SIGN、CD1a、BDCA-2、CD83、HLADR、IDO表达水平,并进行体视学分析与统计。结果正常睾丸及睾丸原发性淋巴瘤患者睾丸组织中均有CD11c、DC-SIGN、CD1a、BDCA-2、HLA-DR、IDO阳性细胞表达;其中CD1a、BDCA-2、HLA-DR、IDO阳性细胞表达分布于睾丸间质区域;CD11c及DC-SIGN在睾丸间质区域及生精细胞中均可见阳性表达;睾丸原发性淋巴瘤患者睾丸组织中CD11c、DC-SIGN、CD1a、HLADR、IDO表达水平较正常睾丸显著增高,且差异有统计学意义(P<0.05);正常睾丸及睾丸原发性淋巴瘤患者睾丸组织中均未见CD83阳性细胞表达。结论睾丸原发性淋巴瘤患者睾丸组织中CD11c+髓样DCs(mDCs),DC-SIGN+间质型DCs,CD1a+DCs、HLA-DR、IDO表达水平显著增高,提示睾丸原发性淋巴瘤患者睾丸组织中表现为耐受性DCs,并在睾丸免疫环境失稳态及肿瘤细胞免疫豁免过程中起重要作用。 Objective To study the characteristic phenotype of dendritic cells (DCs) and subsets in the testicular tissues of primary lymphoma of testis (PLT) by using immunohistochemical (IHC) staining assay.Additionally,we studied the relationship between the functional status of DCs and PLT pathologic characteristics.Method In this research,we compared the expression intensity of DCs specific surface markers and intracellular cytokines CD1a,CD11c,DC-SIGN,BDCA-2,CD83,HLA-DR,IDO in 7 PLT and 10 normal testicular tissues.Results CD11c^+ ,DC-SIGN^+ ,CD1a^+ ,BDCA-2^+ ,HLA-DR^+ and IDO^+ cells were found in normal and PLT testicular tissues and most of them were located in the interstitial tissue of testis.Furthermore,CD11c and DC-SIGN were also found in the spermatogenic cells of normal testicular tissues.The expression of CD11c,DC-SIGN,CD1a,HLA-DR and IDO in PLT patients were significantly increased compared with normal control (P〈0.05).There were virtually absent CD83^+ cells in normal and PLT testicular tissues.Conclusion The expression of CD11c+myeloid DCs (mDCs),DC-SIGN+ Stromal DCs (sDCs),CD1a+ DCs,HLA-DR,IDO were significantly increased in PLT patients compared with normal controls,and associated with a tolerogenic and immature phenotype,which indicated that DCs in PLT may play an important role in imbalance of the immunologically dynamic environment and immune privilege of cancer cells.
