老年雄性大鼠自体移植幼年睾丸组织后血清睾酮水平及间质细胞的变化

摘要 目的行老年雄性SD大鼠自体移植幼年时睾丸组织后检测睾酮水平及移植部位睾丸间质细胞变化,探讨睾丸组织移植对大鼠睾酮水平的影响。方法雄性1月龄SD大鼠20只,测睾酮水平,行右侧睾丸组织切除并放置-80℃冰箱。检测4月龄SD大鼠睾酮水平,检测24月龄睾酮水平,然后右侧腋部皮下自体移植幼年时冰冻睾丸组织。检测25月龄(即睾丸组织自体移植术后1月)血清睾酮水平,检测26月龄(即睾丸组织自体移植术2月后)血清睾酮水平及变化。检测冰冻睾丸组织移植前睾丸间质细胞情况,移植后1月及2月睾丸间质细胞变化。结果 1月龄SD大鼠睾酮水平为(0.067 0±0.031 81)ng/mL;切除一侧睾丸组织4月龄SD大鼠睾酮水平为(2.695 0±0.293 18)ng/mL;切除一侧睾丸组织24月龄SD大鼠睾酮水平为(1.584 2±0.431 19)ng/mL;切除一侧睾丸组织25月龄SD大鼠自体移植大鼠血清睾酮水为(1.789 5±0.298 99)ng/mL;切除一侧睾丸组织26月龄SD大鼠自体移植大鼠血清睾酮水平为(1.516 8±0.413 82)ng/mL。切除一侧睾丸组织24月龄SD大鼠和26月龄SD大鼠(即睾丸组织自体移植术2月后)两组间差异无统计学意义,其余各组两两比较均有统计学意义。冰冻睾丸组织移植前睾丸间质细胞较多,移植后1月睾丸间质细胞有减少趋势,在2月后睾丸间质细胞绝大多数凋亡。结论老年雄性SD大鼠自体移植幼年时睾丸组织后短期内可以提高睾酮水平,因多种因素长期睾酮水平升高不理想。由多种原因移植部位睾丸间质细胞逐渐凋亡。 Objective To investigate the changes of testosterone level and leydig cells of old male SD rats which received autograft transplantation of infant testicular tissues.Methods The testosterone level of 20 1-month old male SD rats was measured,and the right testicular tissues were removed and placed in a refrigerator at -80 ℃.When the rats were 4-month and 24-month old,the testosterone level was detected respectively.After that,the frozen testicular tissues were transplanted subcutaneously in the right axilla.Serum testosterone level was measured,and the changes of leydig cells were detected 1 month and 2 months after transplantation.Results The average testosterone level was 0.067 0±0.031 81,2.695 0±0.293 18,1.584 2±0431 19,1.789 5±0.298 99,and 1.516 8±0.413 82 ng/mL when the rats were 1,4,24,25 and 26 months old.There was no statistically significant difference between the 24-month and 26-month old rats.There were differences between other groups.Before transplantation,there were a large number of leydig cells,while the number of cells decreased 1 month after transplantation,and most of them were apoptotic 2 months after transplantation.Conclusion Autograft transplantation of infant testicular tissues can improve the level of testosterone in the short term,but the long-term effect is not desirable.The leydig cells are apoptotic gradually due to various reasons
