摘要
目的:探讨细胞外组蛋白对M2型巨噬细胞死亡的诱导作用。方法:不同浓度细胞外组蛋白分别处理M2型巨噬细胞,通过MTT检测0、3和6 h后细胞死亡率,光学显微镜观察细胞形态,流式细胞仪检测细胞凋亡率。结果:不同浓度细胞外组蛋白组M2型巨噬细胞死亡率与对照组比较显著升高(P<0.05),细胞外组蛋白诱导M2型巨噬细胞死亡呈现剂量和时间依赖性。细胞外组蛋白诱导M2型巨噬细胞形态发生明显变化。凋亡抑制剂能够显著抑制细胞外组蛋白对M2型巨噬细胞凋亡的促进作用(P<0.05)。结论:细胞外组蛋白能够促进M2型巨噬细胞死亡,凋亡是其死亡方式,细胞外组蛋白可能与脓毒症的发病有关。
Objective :To investigate the effect of extracellular histone on the death of M2 macrophages. Methods : M2 macrophages was treated with different concentrations of extracellular histones ,and the cell death rate was measured by MTT at 0,3, and 6 h. The morphology of the cells were observed by microscope, and the apoptotic rate of M2 macrophages was detected by flow cytometry. Results:The mortality of M2 macrophages in different concentrations of extracellular histone was significantly higher than that in the control group( P 〈 0.05 ). The extracellular histone - in- duced M2 macrophages death was dose and time dependent. The morphological changes of M2 macrophages induced by extracellular histones were significantly changed. The inhibitor of apoptosis could significantly inhibit the apoptosis of M2 macrophages induced by extracellular histone ( P 〈 0.05 ). Conclusion : The extracellular histone can promote the death of M2 macrophages by inducing apoptosis. The extracellular histone may related to the pathogenesis of sep-
