欧报春上下胚轴再生体系的建立 被引量:3

Establishment of Regeneration System of Epicotyls and Hypocotyls of Primrose(Primula vulgaris)

摘要 以欧报春(Primula vularis)种子上下胚轴为外植体,通过外植体的消毒、丛生芽诱导和增殖、生根、炼苗和移栽等技术的研究,筛选出各阶段的最佳培养基配方,从而建立欧报春的再生体系。结果表明:欧报春种子上胚轴能诱导出不定芽,但下胚轴不能。欧报春上胚轴不定芽诱导最佳培养基为MS+NAA 0.4 mg/L+6-BA 0.6 mg/L,诱导率达21.67%;丛生芽增殖的最佳培养基为MS+NAA 0.6 mg/L+6-BA 0.5 mg/L,诱导率达62.96%;生根最佳培养基为MS培养基,诱导率达95.59%;最佳移栽基质为草炭:珍珠岩=3:1(体积比)的混合基质,移栽成活率可达96.67%。 The epicotyls and hypocotyls of Primula v ulgaris were used as explants. Through the disinfection of the explants, induction and multiplication of adventitious buds, rooting culture, seedling preparation and transplantation, the best medium for each stage were screened out so as to establish the regeneration system of Primula vulgaris. Results showed that epicotyls of Primula vulgaris could induce adventitious buds, but hypocotyls could not. The optimal medium of Primula vulgaris to induce adventitious shoots from epicotyls explants was MS+NAA 0.4 rag/L+ 6-BA 0.6 mg/L, with the induction rate of 21.67%. The best medium for induction of multiple shoots was MS+ NAA 0.6 mg/L+6-BA 0.5 mg/L, and the proliferation rate was 62.96%. The optimal medium for rooting was MS, and the rooting rate was 95.59%. The best transplanting medium was the mixture of peat and petite, with the volume ratio of 3:1, and the survival rate of transplanting could reach 96.67%.
