摘要
目的探讨基质细胞衍生因子SDF-1α及其受体CXCR4在类风湿关节炎(RA)发生、发展中的作用及意义。方法用流式细胞术对50例RA患者和25例健康志愿者外周血CD3+CD4+T细胞、CD3+CD8+T细胞表面CXCR4的表达水平进行检测,用ELISA法检测SDF-1α、IL-6在上述两组外周血中的表达情况。结果 RA患者组外周血中CD3+CD4+T细胞和CD3+CD8+T细胞表面CXCR4表达水平和SDF-1α、IL-6的表达均明显高于健康对照组(P<0.05)。直线相关性分析显示,外周血CXCR4及SDF-1α表达水平与IL-6均存在正相关性(P<0.05)。结论 SDF-1α/CXCR4轴及IL-6在RA患者的发病过程中可能起到了重要作用,并且二者有协同作用。
Objective To study the expression of SDF-1α and CXCR4 in the peripheral blood patients with rheumatoid arthritis (RA). Methods 50 RA patients were recruited and 25 healthy volunteers were enrolled as normal controls.The expressions of CXCR4 on CD3+CD4+T cells and CD3+CD8+T cells were detected by flow cytometry in 50 patients with RA and 25 healthy subjects. The level of SDF1αand IL6 were detected by ELISA.Results The expression of CXCR4 on CD3+CD4+T cells and CD3+CD8+T cells and the levels of SDF-1αand I-6 in peripheral blood of patients with rheumatoid arthritis were significantly higher than those of healthy controls (P<0.05). Linear correlation analysis showed that there was a positive correlation between CXCR4 and SDF-1α expression in peripheral blood and IL-6 (P<0.05).Conclusion SDF-1α/CXCR4 axis and IL-6 maybe play an important role in the pathogenesis of rheumatoid arthritis.
