摘要
目的探讨RhoA在卵巢癌细胞SKOV3/DDP耐药过程中的作用。方法应用siRNA干扰技术沉默SKOV3/DDP细胞中RhoA表达,MTT、细胞周期、细胞凋亡、Western Blot检测细胞表型的变化及可能存在的作用机制。结果 Western blot结果表明SKOV3/DDP细胞中RhoA表达水平高于SKOV3细胞。MTT结果提示SKOV3/DDP干扰RhoA后,其对顺铂的药物敏感性上升,细胞周期及凋亡检测发现siRhoA之后导致G2/M期阻滞,细胞凋亡增加。Western blot结果表明沉默RhoA表达后ABCG1以及凋亡相关蛋白Bcl-2、survivin蛋白表达水平明显下降。结论 RhoA通过调节耐药相关蛋白ABCG1以及凋亡相关蛋白Bcl-2,survivin参与卵巢癌细胞耐药。
Objective To investigate the role of RhoA in ovarian cancer cell SKOV3/DDP drug resistance. Methods After siRNA RhoA interference,MTT assays was used to investigate the inhibition rate of SKOV3/DDP after exposure to different concentrations of cisplatin,Western blot assay was used to detect relative drug resistance and apoptosis related protein expression. Results RhoA expression was higher in SKOV3/DDP cell line that SKOV3 cell line. The results of MTT showed that the sensitivity of ovarian cancer cells to cisplatin was increased after RhoA downregulation by siRNA RhoA transfection,besides,RhoA downregulation also induced G2/M phase arrest,and cell apoptosis,inhibited ABCG1,Bcl-2 and survivin protein expression. Conclusion RhoA involves in ovarian cancer cell durg resistance through modulating ABCG1,Bcl-2 and survivin.
